摘 要:[目的]建立一种快速、准确检测物体表面清洁度的方法。[方法]以ATP生物发光微生物检测技术为指导,提取重组的萤火虫荧光素酶并配制成荧光反应试剂,测定其与ATP发光反应值的线性关系;根据ATP含量与细菌数量成正比例关系原理,测定物体表面ATP发光强度值,同时用国标平皿计数法测定其菌落总数,两者对比分析。[结果]ATP生物发光法检测的相对荧光值与物体表面的菌落总数存在良好的线性相关性,利用ATP发光法实时检测细菌总数可以很好地弥补国标法的不足。[结论]ATP生物发光法与传统的细菌总数国标平板计数法相比较具有操作简便、快速灵敏、成本低廉、绿色环保等优点,可以快速、准确地检测物体表面清洁程度。[ Objective ] To establish a rapid and accurate detection method for object surface cleanliness. [ Method ] With ATP bioluminescent microbial detection technology as the guidance, recombinant firefly luciferase was extracted in order to prepare fluorescence reaction reagent. Its linear relation with ATP response value was detected. According to the principal of positive correlation between ATP content and bacterial population, ATP luminous intensity of object surface was detected. Total bacterial count was calculated by national standard plate counting method. And comparative analysis was carried out. [ Result] There was good linear relationship between relative fluorescence unit and total bacterial count of object surface. Real-time detection of total bacteria by ATP bioluminescent method could make up for the disadvantages of national standard. [ Conclusion] Compared with traditional national standard plate counting method, ATP bioluminescent microbial detection technology is more simple, rapid, sensitive and cheap. And it can rapidly and accurately detect the cleanliness of object surface.
