摘　　要：目的观察外周髓鞘蛋白22（PMP22）在涎腺腺样囊性癌（SACC）组织和细胞系中的表达，揭示PMP22与SACC高转移性、神经侵犯性及预后之间的关系。方法收集41例SACC患者的标本，采用免疫组化SP法检测PMP22的表达。以SACC高转移和非转移细胞系SACC—LM和SACC-83细胞为研究对象，利用Western blot检测PMP22的表达差异。结果癌旁正常组织中PMP22表达明显高于SACC癌组织，具有统计学差异（P=0．000）；PMP22在T4期SACC患者中表达阳性率明显低于T1-3期，具有统计学差异（P=0.014）；伴有神经侵犯的SACC患者中PMP22表达阳性率高于无神经侵犯者，具有统计学差异（P=0．038）；3种组织类型的SACC比较，PMP22的表达阳性率也存在统计学差异（P=0．017）；复发组表达阳性率低于不复发组（P=0.003）；不同年龄、性别、肿瘤部位、淋巴结转移等的SACC患者之间PMP22表达阳性率均无统计学差异（P=0．632、0790、0458、0．692）。Western blot显示。PMP22在SACC—LM中表达高于SACC-83（P=0．000）。结论PMP22的表达与SACC的发生、发展及复发可能存在密切联系，可以用于对患者预后的评估。Objective To investigate the expression of PMP22 in salivary gland adenoid cystic carcinoma （SACC） tissue and cell lines and its clinical significance. Methods The expression of PMP22 was examined with immunohistochemistry （SP method） in 41 tissue samples from patients with SACC admitted in Zhejiang Provincial Cancer Hospital from 2002 to 2013. The expression of PMP22 was detected by Western blot in metastatic SACC cell line SACC-LM and non-metastatic SACC cell line SACC-83. Results The expression of PMP22 in adjacent normal tissue was higher than that in SACC tissues （P=0.000）; the expression of PMP22 in T4 SACC tissue was lower than that in T1-3 SACC tissue （P=0.014） ; the expression of PMP22 in nerve invasion group was higher than that in non-nerve invasion group （P=0.038）; the expression of PMP22 in three different histological types of SACC was significantly different （P=0.017）; the expression of PMP22 in recurrent group was lower than that in non-recurrent group（P=0.003）. The expression of PMP22 was not correlated with age, sex, tumor Iocaton and lymph node metastasis （P=0.632, 0.790, 0.458, 0.692）. Western blot showed that PMP22 level in SACC-LM were significantly higher than that in SACC-83 cells. Conclusion The expression of PMP22 is closely related to the occurrence and development of SACC, and it may have prognostic value for SACC.
