摘  要:目的:探讨番茄红素对体外培养的人非小细胞肺癌H520细胞增殖、侵袭和迁移等生物学行为的作用。方法:采用MTT法检测不同浓度番茄红素对H520细胞作用24、48及72小时后细胞的存活率,Transwell小室实验检测7μmol/L番茄红素对H520细胞侵袭能力的影响,划痕试验检测7μmol/L番茄红素对H520细胞迁移能力的影响。实时定量PCR检测不同浓度番茄红素作用于H520细胞24小时后金属基质蛋白酶(MMP)-2和-9的表达变化。结果:番茄红素能够抑制人非小细胞肺癌H520细胞的增殖,呈时间和剂量依赖性(P<0.05)。同时番茄红素减少侵入Transwell系统下室的细胞数目(P<0.05)并抑制H520细胞向划痕区域的迁移距离(P<0.05)。番茄红素剂量依赖性地降低细胞中MMP-2和MMP-9的表达(P<0.05)。结论:番茄红素能够抑制H520细胞的增殖,并且通过抑制MMP-2和-9的表达抑制其侵袭和迁移能力。Objective: To investigate the effects of lycopene on the proliferation,invasion and migratory ability of human non- small cell lung cancer H520 cells. Methods: The survival rate of H520 cells treated with varied dose of lycopene for 24,48 and 72 h was detected by MTT assay. Transwell and wound healing analysis were adopted to evaluate invasion and migratory ability of H520 cells stimulated by 7μmol / L of lycopene,respectively. mRNA expression of matrix metalloproteinase( MMP)- 2 and- 9 was examined by real- time PCR. Results: Lycopene inhibited the proliferation of H520 cells in a time- and dose- dependent manner( P〈 0. 05). 7μmol / L of lycopene reduced the number of H520 cells invaded into the lower chamber of Transwell system( P〈 0. 05) as well as the distance that cell migrated to heal the wound. Lycopene inhibited the mRNA expression of MMP- 2 and- 9 in a dose dependent manner in H520 cells. Conclusion: Lycopene inhibited the proliferation,invasion and migratory abilities of H520 cells probably through interfering the expression of MMP- 2 and MMP- 9.
