摘 要:目的利用沙门菌分子分型技术,分析不同来源的菌株之间的遗传相关性,通过同源性分析追溯新疆某县一起食物中毒发生的原因。方法对样品进行培养获得3株疑似沙门菌。通过脉冲场凝胶电泳(PFGE)技术对菌株进行鉴定及分子溯源。结果 5份患者粪便标本中有2份分离到菌株,3份食品样品中有1份分离到菌株。3株菌株初步生化鉴定结果为TSI(K/A)、产气、H2S:+;赖氨酸脱梭酶:+;血清学分型结果相同,均为O:9:12:+;H:g:m:+;H2相:-,鉴定为肠炎沙门菌。药敏检测结果显示对四环素等8种药物100%敏感,对萘啶酸100%耐药。PFGE分子分型及同源性分析,3株肠炎沙门菌的DNA图谱完全相同。结论 3株肠炎沙门菌株的DNA图谱100%同源,确认为一起由肠炎沙门菌污染了食品而引起的食物中毒。Objective To clarify of genetic correlation of strains from different isolates using molecular technologies; to define the causal agent of food poisoning in a county of Xinjiang province using homology analysis. Methods Bacteria strains were cultured for obtaining 3 Salmonella- like strains. Strains identification and molecular traceability were conducted by pulsed-field gel electrophoresis( PFGE). Results Two strains were isolated from five specimens of patients stool,and one strain was obtained from food samples. The preliminary biochemical identified results were as follows: TSI( K/A),gas,H2S: +,lysine decarboxylase: +; the results of serotyping were same: O: 9: 12: +; H: g: m: +; H2 phase:-. The identification result was Salmonella enteritidis. Susceptibility testing showed 100% sensitivity of eight kinds of tetracycline drugs,100% of nalidixic acid- resistant. With molecular typing and homology analysis of PFGE,three Salmonella enteritidis showed the same DNA fingerprinting. Conclusion The DNA fingerprinting of the three strains of Salmonella enteritidis was with 100% homology,which indicated the causal agent of food poisoning was identified as contamination caused by Salmonella enteritidis.
