摘 要:为了找到鬼针草叶蛋白提取最佳生长期和有效提取方法,研究了鬼针草不同生育期和不同方法对叶蛋白提取的影响。结果表明:在鬼针草4个不同生育期中采用直接加热法提取,叶蛋白提取率和沉淀物中粗蛋白含量依次降低,营养生长期(20.2 mg/g,61.36%)>现蕾期(15.9 mg/g,61.25%)>盛花期(14.7mg/g,60.97%)>结果期(12.1mg/g,54.22%),均以营养生长期为最高。在鬼针草的营养生长期,采用多种方法提取叶蛋白,叶蛋白提取率:酸化加热+聚乙二醇沉淀法(29.4mg/g)>酸化加热+乙醇沉淀法(27.9mg/g)>酸化加热+氯化钠法(24.7mg/g)>酸化加热法(23.1mg/g)>直接加热法(20.2mg/g);沉淀物中粗蛋白含量:酸化加热+乙醇沉淀法(65.35%)>酸化加热+氯化钠法(60.72%)>酸化加热+聚乙二醇沉淀法(57.13%)。The effect of different B.bipinnata growth stage and different extraction methods on leaf protein extraction rate fromB.bipinnata was studied to probe the optimum growth stage of B.bipinnata for extracting its leaf protein and effective extraction method.Results:The leaf protein rate and crude protein content in the sediment at vegetative,budding,full flowering and fruit-bearing stage are20.2mg/g and 61.36%,15.9 mg/g and 61.25,14.7 mg/g and 60.97%,and 12.1 mg/g and 54.22%respectively by using the direct heating method.When B.bipinnatais at the vegetative stage,the leaf protein extraction rate of the acidification heating + polyethylene glycol,acidification heating+ ethanol,acidification heating + NaCl,acidification heating and direct heating method is 29.4 mg/g,27.9 mg/g,24.7 mg/g,23.1 mg/g and 20.2 mg/g respectively but the crude protein content in the sediment of acidification heating + ethanol,acidification heating + NaCl and acidification heating + polyethylene glycol method is 65.35%,60.72% and 57.13%respectively.
