摘  要:目的探讨大蒜素对宫颈癌He La细胞周期的影响及其相关分子机制。方法不同浓度大蒜素及不同时间作用于体外培养的He La细胞,采用WST-8法测定大蒜素对He La细胞增殖的影响;流式细胞术测定大蒜素对He La细胞周期的影响;不同浓度大蒜素处理He La细胞24h后,用逆转录-聚合酶链反应(RT-PCR)法和蛋白印迹(Western blot)法分别检测β-catenin mRNA及蛋白表达。结果大蒜素以浓度和时间依赖的方式抑制He La细胞的增殖,阻遏细胞周期进展,下调关键蛋白β-catenin在胞质的表达。结论大蒜素抑制Wnt信号的活化,下调关键蛋白β-catenin在胞质的表达,从而阻遏细胞周期进展,抑制He La细胞增殖,为宫颈癌防治新药的开发提供理论依据。Objective To investigate the effects of garlicin on the cycle of cervical cancer He La cells and its molecular mechanism. Methods He La cells cultured in vitro were given different concentrations of garlicin on different time. The effects of garlicin on the proliferation of He La cells were measured by WST-8 method,while the effects of garlicin on the cycle of He La cells were measured by flow cytometry. After being treated by different concentrations of garlicin for 24 hours,the expression of mRNA protein and β-catenin of He La cells were respectively measured by reverse transcriptase polymerase chain reaction( RT-PCR)method and Western blot( Western blot) method. Results Garlicin inhibited the proliferation of He La cells,and repressed the progress of cell cycle and the expression of key protein β-catenin in the cytoplasm. Conclusion Garlicin inhibited the activation of Wnt signal,repressed the expression of key protein β-catenin in the cytoplasm,and thereby prevented the progress of cell cycle and inhibited the proliferation of He La cells,which provided a theoretical basis for the development of new drugs to prevent and treat cervical cancer.
