摘  要:研究纤维素酶提取五味子多糖的工艺及体外抗氧化活性。在单因素试验的基础上以响应面分析法优化提取工艺,最佳提取工艺条件为:酶解温度65℃,酶解时间为2.98 h,提取液p H 7.98,酶用量2.06%,料液比1︰40(g/m L),药材粒径80目,五味子多糖的提取率为19.38%。对五味子多糖的体外抗氧化活性进行研究。结果显示,五味子多糖体外总抗氧化能力和还原力显著,与浓度呈明显的量效关系,对DPPH自由基,羟自由基,超氧阴离子自由基具有较强的清除能力。To study the cellulase-assisted extraction technology of polysaccharides from Schisandra chinensis Baill and its antioxidant activity. On the basis of single extraction experiment, response surface methodology(RSM) was used to optimize the extraction technology. The optimum conditions was as follows: enzymolysis temperature of 65 ℃, enzymolysis time of 2.98 h, p H 7.98, amount of cellulase of 2.06%, liquid to solid ratio of 1︰40(g/m L), sample particle of 80 meshes. Under these optimum conditions, the extraction efficiency of S. chinensis polysaccharides has reached 19.38%. The antioxidant activity was also studied, results showed that polysaccharides posed significant antioxidant activity in vitro, the scavenging actives on total antioxidant capacity, reducing power, DPPH radical, hydroxyl radical and superoxide radical increased with increasing of polysaccharides concentration.
