摘  要:运用电子顺磁共振(EPR)和细胞增殖检测试剂盒(MTS)两种方法研究4个双羟基香豆素化合物及天然产物V_C的清除DPPH自由基及抗肿瘤细胞增殖的能力。结果表明,香豆素化合物清除DPPH自由基EC_(50)值和抗肿瘤细胞增殖活性IC_(50)值的范围分别为9.04μmol/L^37.32μmol/L和21.76μmol/L^38.48μmol/L,均优于V_C自由基清除能力(EC_(50)为42.44μmol/L)和抗肿瘤细胞增殖能力(IC_(50)为>100μmol/L)。其中,化合物1a的清除自由能力最强(EC_(50)为9.04μmol/L),化合物2b的抗肿瘤活性最高(IC_(50)为21.76μmol/L)。因此,与V_C相比,4个双羟基类化合物表现出较强的自由基清除能力和抗肿瘤细胞增殖活性,为具有癌症防治功能的天然产物抗氧化剂的开发利用提供理论基础和试验依据。The free radical DPPH scavenging and anti-tumor activity of four dihydroxy coumarin derivatives and VC were investigated using electron paramagnetic resonance(EPR) and MTS methods. The results showed that the ranges of EC50(radical scavenging ability) and IC50(antitumor activity) values of four compounds were 9.04 μmol/L-37.32 μmol/L and 21.76 μmol/L-38.48 μmol/L, respectively, and therefore the radical scavenging and anti-tumor ability of four compounds were more potential than those of VC with EC50=42.44 μmol/L and IC50100 μmol/L, respectively. Compound 1a(EC50=9.04 μmol/L) and 2b(IC50=21.76 μmol/L) were the most active compounds at DDPH radical scavenging and antitumor, respectively. In conclusion, compared with VC,four coumarin compounds exhibited more potential ability at radical scavenging and antitumor cell proliferation,and were considered as the natural antioxidant with free radical scavenging abilityand antitumor activity.
