摘  要:以别隐品碱提取率为指标,先考察提取溶剂加酸和酶解预处理对两面针超声提取的适用性,再用正交试验优化超声功率、提取次数和溶剂量,最后动态过程优化超声提取时间。结果证明提取溶剂p H≤3不利于别隐品碱的提取。最佳工艺条件为:复合酶预处理后,以体积分数60%乙醇超声(250 W)提取3次,第1次以7倍量溶剂提取18 min,第2次以3倍量溶剂提取12 min,第3次以2倍量溶剂提取12 min,别隐品碱提取率为89.5%。该工艺经济、高效、节能、省时,可为开发利用别隐品碱提供实验基础。In this study, the ultrasonic-assisted extraction technology for allocryptopine from Zanthoxylum nitidum was in- vestigated. Extraction with acidic solvent and pretreatment performance with enzymes were compared. The effects of ultra- sonic power,times of extraction and solvent volume on extraction performance were optimized by orthogonal test. Extrac- tion duration was optimized in dynamic process, with extraction yield of allocryptopine as the evaluation index. The re- sults showed that the extraction yield of allocryptopine with neutral solvent was better than that with acidic ( pH ≤3 ) sol- vent. The optimal extraction process was as foUows:Z, n/t/dum powder was pretreated with ceUulase ( 1: 250) and pecti- nase ( 1: 250) for 30 min in 3 times volume of acetic acid-sodium acetate buffer ( pH 5) at ambient temperature ( ≥ 30 ℃ ) ,and then extracted 3 times by ultrasonic wave (250 W) with 60% ethanol as solvent,extracted 18 rain with 7 times solvent at the first time,extracted 12 min with 3 times solvent at the second time, extracted 12 min with 2 times solvent at the third time. Under the optimized extraction procedure,the extraction yield of allocryptopine was 89.5%. The opti- mized extraction process is economic, efficient, energy and time saving. It provides experimental basis for industrial pro- duction.
