摘  要:本文探索和优化一种高效的复合酶解技术从夏枯草中提取多糖。在单因素试验的基础上,选择酶添加量(A)、提取温度(B)和溶液p H值(C)三因素三水平的Box-Behnken Design(BBD)试验设计,响应面优化提取工艺。得到最佳提取条件:纤维素酶浓度0.4%、果胶酶浓度0.8%、木瓜蛋白酶浓度0.8%,提取温度61℃,p H 6.2。在此条件下夏枯草粗多糖的提取得率为11.57±0.48%(n=3),与预测得率(12.01%)基本一致。说明优化得到的工艺合理、可行,复合酶提取得率高、能耗低,是一种很有希望的提取方法。In this study,an efficient complex enzyme extraction( CEE) technology was developed and optimized to extract polysaccharides from Prunella vulgaris. On the basis of single factor test,response surface methodology( RSM) based on a three- factor three-level Box- Behnken Design( BBD) was employed to optimize the extraction conditions including enzyme concentration,extraction temperature and p H. The optimal extraction conditions were as follows: cellulase 0. 4%,pectinase 0. 8%,papain 0. 8%,extraction temperature 61℃,p H 6. 2. Under these conditions,the experimental yield was 11. 57 ± 0. 48%,which is well in close agreement with the value predicted( 12. 01%) by the model. These results demonstrated that the optimal extraction technology is reasonable and feasible.The CEE is a promising extraction method that offers higher yield and lower energy cost.
