摘 要:目的:研究黑曲霉真菌诱导子对青钱柳悬浮培养细胞合成三萜的影响。方法:采用植物细胞培养技术,建立稳定的青钱柳细胞悬浮培养体系,以外源黑曲霉诱导子刺激细胞合成三萜,分析诱导子浓度、添加时间及持续作用时间对细胞生长和三萜合成的影响。结果:在稳定期初期(第6天)添加50-300μg/m L的黑曲霉诱导子对细胞生长基本没有影响,但都能刺激细胞合成三萜,200μg/m L的刺激效果最好;在细胞生长周期的不同时间点添加200μg/m L的诱导子,对细胞生长和三萜合成的促进效果不一样,第0天(即接种时)添加对细胞生长的抑制作用最大,对数期中期(第4天)添加时细胞生物量比对照组降低了7.83%,但三萜含量却提高到对照组的9.10倍,对数期中期添加促进三萜合成效果最好;添加诱导子作用3 d后收获细胞,三萜含量和产量均达到最高,分别是对照组的10.79倍和10.21倍。结论:黑曲霉诱导子能够有效地促进青钱柳悬浮细胞合成三萜。Objective:In order to study the effect of Aspergillus Niger(A.niger) inducer on the biosynthesis of triterpenoids in suspended cultured Cyclocarya paliurus cells.Methods:A stable Cyclocarya paliurus suspension culture was established.An A.niger inducer was used to stimulate the triterpenoid synthesis,and the concentration,the adding time and the duration time of this inducer were analyzed.Results:50-300 μg/m L A.niger inducers almost had no inhibition on the cell growth,but stimulated the synthesis of triterpenoids when the inducers were added into the cell cultures at the early stable period(the 6th day),and 200 μg/m L was the optimized concentration.Cell biomass and triterpenoid production were different as the inducer was added in different period.The lowest cell biomass was recorded when a200 μg/m L A.niger inducer was added on the 0th day(in cell seeding).When the inducer was added on the mid-log phase(the 4th day),the cell biomass decreased 7.83%,but the content of triterpenoids increased 9.10 times compared with the control,the 4th day was the best adding time.The content and yield of triterpenoids both peaked after 3 day inducement of a 200 μg/m L A.niger inducer,which were 10.79 times and 10.21 times higher than that of the control,respectively.Conclusion:The A.niger inducer stimulated suspension-cultured Cyclocarya paliurus cells to synthesize triterpenoids effectively.
