摘  要:目的探讨血清循环DNA微卫星位点杂合性缺失(LOH)对宫颈癌与卵巢癌的诊断应用价值。方法选取6个微卫星位点,采用血液基因组DNA提取试剂盒抽提各组血清标本循环DNA、PCR、变性聚丙烯酰胺凝胶电泳及硝酸银染色对63例宫颈癌和33例卵巢癌(病例组)、63例子宫肌瘤(良性对照组)、63名健康成年女性血清标本(健康对照组)进行检测。结果宫颈癌在D3S1038、D3S1300、D3S1228位点LOH频率分别为53.4%、60.3%、57.1%;卵巢癌在D17S579、D17S855、D17S786、D3S1038位点的LOH频率分别为:45.5%、42.4%、27.3%、51.3%;所有位点的病例组与健康对照组的LOH差异有统计学意义(P<0.05);宫颈癌D3S1228位点与良性对照组的LOH差异有统计学意义(P<0.05);卵巢癌D17S579与D17S855位点与良性对照组的LOH差异有统计学意义(P<0.05)。结论宫颈癌D3S1228位点及卵巢癌D17S579、D17S855位点具有较高的LOH,为进一步研究血清循环DNA作为宫颈癌与卵巢癌的遗传学辅助诊断指标提供了一定的实验基础。Objective To explore the efficiency of serum circulating DNA microsatellite LOH in the diagnosis of cervical cancer and ovarian cancer.Methods Six sites of microsatellite were selected.Serum circulating DNA(cDNA)was extracted with blood genomic DNA extraction kit.LOH were detected with polymerase chain reaction(PCR)and denaturing polyacry1 amide gelelectrophoresis in 63 cases of varian cancer,33 cases of ovarian cancer(pathological group),63 cases of hysteromyoma(benign group)and 63 healthy adult women serum(healthy control group).Results The rates of LOH on cervical cancer at D3S1038,D3S1300 and D3S1228were 53.4%,60.3% and57.1% respectively.The rates of LOH on ovarian cancer at D17S579,D17S855,D17S786 and D3S1038 were45.5%,42.4%,27.3% and 51.3%respectively.Significant difference was observed in LOH between pathological group and healthy control group at all the sites(P〈0.05).There was significant difference in LOH rates between benign group and cervical cancer at D3S1228,and between benign group and ovarian cancer at D17S579 and D17S855(P〈0.05).Conclusion LOH on D3S1228 are common in cervical cancer,and LOH on D17S579 and D17S855are common in ovarian cancer.This study could provide the experimental basis for further study on serum c-DNA as auxiliary diagnostic indicators of cervical cancer and ovarian cancer.
