摘  要:目的:研究库拉索芦荟在黄曲霉毒素B_1(aflatoxin B_1,AFB_1)致大鼠急性肝损伤中的干预作用及机理。方法:采用单次灌胃2.0 mg/kg(以体质量计)AFB_1建立大鼠急性AFB_1肝损伤模型,通过体质量、肝功能指标、肝组织病理学变化评价库拉索芦荟的干预效果,并通过测定肝组织脂质过氧化产物、各抗氧化物水平,探讨库拉索芦荟拮抗急性AFB_1肝毒性的作用机理。结果:库拉索芦荟具有显著降低急性AFB_1肝毒性的功效,能有效抑制急性AFB_1中毒大鼠体质量下降,降低血清谷丙转氨酶(glutamate pyruvate transaminase,GPT)、谷草转氨酶(glutamic oxalacetic transaminase,GOT)和碱性磷酸酶(alkaline phosphatase,ALP)活性以及总胆红素(total bilirubin,TBIL)含量;同时能显著改善肝脏组织病变情况,降低丙二醛(malondialdehyde,MDA)生成量,提高还原型谷胱甘肽(reduced glutathione,GSH)含量以及谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)、超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽-S-转移酶(glutathione-S-transferases,GST)活力,增强机体抗氧化及解毒能力。Objective: To investigate the possible hepatoprotective activity of Aloe vera against aflatoxin B_1(AFB_1)-induced liver damage and its underlying mechanisms. Methods: The acute AFB_1-intoxicated rat model was established by intragastric administration of AFB_1 at the dose of 2.0 mg/kg. The hepatoprotective effect of Aloe vera on AFB_1-intoxicated rats was evaluated by measuring body weight, serum markers and histopathological examination. And the mechanisms involved were elucidated by measuring the levels of lipid peroxidation and antioxidan ts in livers. Results: The treatment of Aloe vera effectively inhibited the body weight loss in AFB_1-intoxicated rats, and significan tly reduced serum glu tamate pyruvate transaminase(GPT), glutamic oxalacetic transaminase(GOT), alkaline phosphatase(ALP), and total bilirubin(TBIL) levels. In addition, Aloe vera was shown to be effective in improving hepatic histopathological changes, lowering malondialdehyd e(MDA), and elevating the levels of reduced glutathione(GSH), gl utathione peroxidase(GSH-Px), superoxide dismutase(SOD) and glutathione-S-transferase(GST). These results suggest that Aloe vera can remarkably enhance the antioxidant and detoxifying capacities of AFB_1-intoxicated rats, and possesses a potent protective effect against AFB_1-induced hepatotoxicity.
