摘 要:采用正交实验筛选东紫苏总黄酮的提取工艺;利用清除超氧阴离子自由基(O_2^-·)和清除双氧水(H_2O_2)实验,考察东紫苏总黄酮的抗氧化活性;通过小鼠体内碳粒廓清实验,评价东紫苏总黄酮的免疫活性。结果显示,东紫苏总黄酮的最佳提取工艺为:料液比1∶20(g/m L)、提取温度60℃、回流时间3 h;在50~200μg/m L的浓度范围内,东紫苏总黄酮对O_2^-·和H_2O_2的清除率分别为26.36%~73.94%和48.56%~77.39%,半数抑制浓度IC_(50)分别为108.07μg/m L和50.59μg/m L;中剂量和高剂量组可显著提高小鼠碳粒廓清指数K和吞噬指数α。表明东紫苏总黄酮具有良好的抗氧化和免疫活性。The extract conditions of total flavonoids from Elsholtzia bodinieri Vaniot were optimized by designing orthogonal test.Its antioxidant activity was assessed thorugh the superoxide anion radical and hydrogen peroxide scavenging ratio.Its immune activity was researched by carbon clearance test.The optimal extraction conditions were determined as follow: the extraction temperature was 60 ℃,the reflux time was 3 h,the liquid- material ratio was 1∶ 20( g / m L).At the range of concentration of 50 - 200 μg / m L,the scavenging ratio for O2^-· and H2O2 was26.36% -73.94% and 50.56% - 77.39%,and IC50 was 108.07 μg / m L and 46.53 μg / m L,respectively.The carbon clearance index K and phagocytic ratio α were increased significant in the high- dose group and middle- dose group.Thus,total flavonoids from Elsholtzia bodinieri Vaniot possessed good antioxidant and immune activities.
