摘 要:为获得一种高效、快速的筛选高产3-羟基丁酮菌株的方法,首先对诱变后的34株突变株分别进行摇瓶发酵和48孔板发酵,验证两种方法之间的相关系数R为0.806 2,表明两种方法具有较好的一致性,48孔板可以用于产3-羟基丁酮菌株的大批量筛选。然后采用响应面分析法对48孔板发酵培养基组分进行优化。结果表明,在优化的条件下,3-羟基丁酮产量达到41.17 g/L,比优化前提高了30.08%,且3-羟基丁酮转化率由原来的27.31%提高到33.12%。In order to obtain an efficient, fast method for screening strains with acetoin high-yield, 34 mutant strains obtained by mutagenesis were cultivated in shake flask and 48-well microplate fermentation, respectively. The correlation coefficient of the two fermentation methods was 0.806 2, which showed that they had a good consistency, and the 48-well microplate could be used for mass screening acetoin-producing strains. The fermentation medium components of48-well microplate were optimized by response surface methodology. The results showed that under the optimum conditions, the acetoin yield was up to 41.17 g/L, which was 30.08% higher than that before optimization, and the conversion of acetoin increased from 27.31% to 33.12%.
