摘 要:[目的]以乙酸酐和芳樟醇为底物,采用脂肪酶催化法合成乙酸芳樟酯。[方法]通过考察不同种类的脂肪酶对该反应的催化效果,研究不同种类固定化载体对该脂肪酶活性的影响,探讨固定化后的脂肪酶在有机相中催化合成乙酸芳樟酯的条件。[结果]获得了一种对该反应催化效率较高的脂肪酶Novozym 435,得出其最佳固定化条件,确定了固定化后的脂肪酶在有机相中催化合成乙酸芳樟酯的最佳反应条件:异辛烷为最佳有机溶剂,以摩尔比为1∶1.5的芳樟醇和乙酸酐为底物,底物浓度为0.5 mol/L,在45℃下振荡反应24h,此条件下乙酸芳樟酯的收率较高。[结论]该反应体系的建立为今后工业生物催化法合成乙酸芳樟酯奠定了良好的基础。[Objective] To synthesize linalyl acetate by catalyzing substrate of acetic anhydride and linalool. [Method] Through investigating the catalysis effect of different kinds of lipases,the effect of different kinds of fixed carriers on the lipase activity were studied,the conditions in which the ester of acetic anhydride and linalool was catalyzed by the fixed lipase in organic solvent were explored. [Result] A lipase Novozym 435 which can catalyze the reaction most efficiently was obtained,as well as the best conditions for the fixation. The best conditions for the reaction were as follows: the best organic solvent must be isooctane; substrate of acetic anhydride and linalool should be 1∶ 1. 5 by Mole;the concentration of the substrate must be 0. 5 mol / L; the temperature should be 45 ℃; shaking reaction for 24 h. Under these conditions we can get linalyl acetate as much as possible. [Conclusion]The reaction system lays a solid foundation for synthesizing linalyl acetate in the bioindustry area in the future.
