摘  要:以黑曲霉TJ02为出发菌,对其孢子进行硫酸二乙酯化学诱变,筛选得到遗传稳定的β-葡萄糖苷酶高产菌株DES-7。诱变株DES-7产酶能力可达28 IU/mL以上,较出发菌株提高30%。同时,对该菌株的发酵培养基进行优化,以玉米芯为碳源,酵母粉和硫酸铵为氮源,其产酶能力达到39 IU/mL,较优化前提高了39.3%。此外,对该菌株的β-葡萄糖苷酶的最适温度和pH以及温度稳定性和pH稳定性进行了测定。Based on the strain of Aspergillus niger TJ02, a β-glucosidase high-yield mutant DES-7 with genetic stability was obtained by diethyl sulfate mutagenisis. Compared with the original strain, the enzyme yield of strain DES-7 up to 28 IU/mL with 30% increasement. Meanwhile, the components of fermentation medium were optimized and the enzyme production yield reached 39 IU/mL with 39.3% increasement, taking corncob and yeast powder and ammonium sulfate as carbon source and nitrogen source, respectively. Besides, the optimal temperature and pH, the stability of temperature/pH of the β-glucosidase were also determined.
