摘  要:目的探讨非小细胞肺癌(Non-Small Cell Lung Cancer,NSCLC)组织中Raf激酶抑制蛋白(Raf Kinase Inhibitor Protein,RKIP)和果蝇Zeste基因增强子人类同源物(Enhancer of Zeste Homolog,EZH2)的表达及其与临床病理特征的关系。方法应用免疫组织化学方法检测84例肺癌组织及相应的癌旁正常肺组织中RKIP与EZH2蛋白的表达情况,分析两者与肺癌临床病理学参数的关系。结果肺癌组织中RKIP的阳性表达率为36.90%(31/84),低于癌旁正常肺组织63.10%(53/84),差异有统计学意义(χ2=11.524,P<0.05);EZH2的阳性表达率为60.71%(51/84),高于癌旁正常肺组织39.29%(33/84),差异有统计学意义(χ2=7.714,P<0.05)。其表达水平与肺癌的分化程度、TNM分期、淋巴结转移及术后生存时间有明显关系;RKIP蛋白与EZH2蛋白的表达水平呈负相关(r=-0.446,P<0.05)。结论RKIP与EZH2在肺癌的发生、发展中起重要作用,RKIP的低表达以及EZH2的过表达,促进了肺癌细胞的侵袭和转移。Objective This work was to investigate the expression of raf kinase inhibitor protein (RKIP) and the enhancer of zeste homolog (EZH2) in lung carcinoma and to explore its correlation with the clinicopathologic features. Methods The expression of RKIP and EZH2 was measured using immunohistoehemistry in 84 lung carcinoma specimens and 84 adjacent normal lung tissue samples. Statistical analysis was used to determine the correlation between their expression and clinicopathologic parameters. Results Statistical analysis revealed that RKIP positive staining rate was 36.90% (31/84) in tumor tissues, which is lower than that of 63.10% ( 53/84 ) in normal lung tissues ( P 〈 0. 05 ). However, EZH2 positive staining rate was 60.71% (51/84) in tumor tissues, significantly higher than that of 39.29% (33/84) in normal lung tissues (P 〈 0.05). The expression of RKIP and EZH2 were significantly correlated with the differentiation of tumor, TNM staging, the development of lymph node metastasis and the survival time ( P 〈 0.05, respectively). There was a negative correlation between the expression of RKIP and EZH2 in lung carcinoma ( r = - 0. 446,P 〈 0.05). Conclusion The expression of RKIP and EZH2 may play an important role in the initiation and development of lung carcinoma. The low expression of RKIP and high expression of EZH2 may promote the invasion and metastasis of lung cancer.
