摘 要:发展了一种高效液相色谱法测定葡萄酒中苯甲酸和山梨酸含量的方法。采用反相C18色谱柱(250mm×4.6mm i.d.,5μm),以甲醇/0.02mol/L乙酸铵(5/95,V/V)为流动相,流速1.0mL/mjn,检测波长230nm,柱温为40℃。实验结果表明:苯甲酸和山梨酸在5.0-100.00mg/L浓度范围内线性相关系数分别为0.9999和1.0,方法检出限分别为0.54mg/L和0.45mg/L(S/N=5),平行5份葡萄酒样品,苯甲酸和山梨酸含量相对标准偏差分别为2.15%和0.21%;加标浓度分别为100mg/L和250mg/L,苯甲酸和山梨酸加标回收率分别为103.90%-108.90%和96.1%-103.9%。该方法定量结果准确,能够满足葡萄酒中苯甲酸和山梨酸含量测定的要求。In this paper, a method for determination of benzoic acid and sorbic acid in grape wine by high performance liquid chromatography(HPLC)was developed. The separation was performed by C18 column (250mm×4.6mm i.d., 5μm) with gradient elution of methanol/0.02mol/L ammonium acetate (5:95,v:v) at 40℃. The flow rate was 1.0 mL/ min and the detection wavelength was 230 nm. The results of benzoic acid and sorbic acid showed that the linear range was from 5.0 to 100.0 mg/L with a linear correlation coefficient of 0.9999 and 1.0, respectively. The limit of quantification was 0.34 mg/L and 0.43 mg/L in grape wine sample (S/N=3). The relative standard deviations of parallel determination (n=5) were 2.13% and 0.21%. The recoveries were 103.90%-108.90% and 96.1%-103.9% of 100.0 mg/L and 250.0 mg/L in grape wine. It was indicated that the method was accurate and repeatable for the determination of benzoic acid and sorbic acid in grape wine.
