摘 要:目的探究在正畸力的作用下大鼠牙周膜张力侧Runx2与β-catenin的表达变化。方法建立大鼠左侧上颌第一磨牙近中移动模型。将36只7周龄健康雄性SD大鼠随机等分为6组,分别为正畸加力12 h、24 h、5 d、7 d、14 d组与不加力组,每组6只。使用一段镍钛拉簧固定于左侧上颌第一磨牙和上颌两个中切牙之间,对第一磨牙施加向近中移动的50 g正畸力值。不加力的大鼠未安装正畸加力装置作为对照组。实验结束后,经常规取材分离上颌骨,取张力侧2 mm牙周膜组织在液氮中研磨。通过实时定量荧光PCR方法检测Runx2和β-catenin。应用SPSS 16.0软件和单因素方差分析方法,定量分析Runx2和β-catenin在大鼠正畸牙齿移动中牙周膜张力侧的表达。结果 Runx2与β-catenin的表达随时间延长而增加。Ru n x 2与β-cateni n在加力7 d后达峰值,14 d后逐渐降低。结论 Ru n x 2与β-cateni n可能参与牙周组织的改建,提示Wnt信号通路可能是参与牙周组织改建的途径之一。Objective To explore the expression levels of Runx2 and β-catenin in periodontal tissue of the tension side during orthodontic tooth movement.Methods A model of OTM on the left maxillary first molar in rats was established.A total of 36 SPF male Sprague-Dawley(SD) rats were randomly divided into six groups:0 h,12 h,24 h,5 d,7 d,and 14 d after orthodontic force application.There were six rats in each group.A Nickel-titanium closed-coil spring was fixed between the maxillary left first molar and incisors,and was used to deliver an initial 50 g mesial force to the left maxillary first molar.The right maxillary molars without force application served as control.The expression changes of Runx2 and β-catenin in periodontal tissue of tension side at different staining period were detected with RTPCR methods.The data were analyzed with SPSS 16.0 software.Results The expression levels of Runx2 and β-catenin increased during orthodontic tooth movement and were related to the movement time.They reached the peak after straining for 7 d and then began to decrease after straining for 14 d.Conclusions Runx2 and β-catenin participated in the reconstruction of periodontal tissue,suggesting that Wnt signaling pathway might be one of regulation pathways in periodontal tissue remodeling during orthodontic tooth movement.
