摘　　要：采用Folin-Ciocalteu法测定金针菇多酚,在单因素基础上,利用二次正交旋转组合设计,优化测定金针菇多酚含量的条件.结果表明,利用Folin-Ciocalteu法测定金针菇多酚含量的最佳条件:添加2mL的福林酚试剂以及2.4mL的15%Na_2CO_3溶液,反应时间为70min,温度45℃,测定波长为730nm.The total polyphenol in Flammulina velutipes was determined by Folin-Ciocalteu method,and the stuitable chromogenic conditions for determination were optmized by quadratic orthogonal rotation combination design which was based on the single-factor experiments. Results showed that the optimum chromogenic conditions were as follows：2 mL Folin phenol reagent,2.4 mL 15% Na2CO3 at 45 ℃ for 70 min,and the detection wavelength was 730 nm.
