摘 要:目的:我国发酵法生产L-色氨酸存在着产酸率、糖酸转化率和提取率较低以及检测速度较慢等不足之处,为了提升发酵控制水平及产物浓度的快速检测,采用近红外检测技术建立发酵过程中L-色氨酸浓度预测模型.方法:利用近红外光谱技术,结合偏最小二乘法,建立发酵液中L-色氨酸浓度预测模型.结果:在光谱预处理为二阶导数、波数范围为6101.8~5450 cm-1条件下获得最优L-色氨酸浓度预测模型.经验证,该模型具有一定的准确性和可靠性,其预测值与测量值仅有5.16%的偏差.结论:该模型具有较好的预测能力,可为大肠杆菌L-色氨酸发酵过程中浓度快速检测提供可靠数据,并为L-色氨酸发酵过程控制提供理论和实践依据.Objective:L-tryptophan fermentation in our country exhibited lower production,conversion ratio and extraction ratio as well as slower detection,in order to improve the fermentation control level and product concentration of rapid detection,L-tryptophan acid concentration prediction model is established in the fermentation process based on near infrared detection technology.Methods:In this report,the calibration model for L-tryptophan concentration monitoring was developed by the near infrared spectroscopy.Results:L-tryptophan concentration prediction model was obtained under the condition of two order derivative and the wave number range region was6101~5450 cm-1.The traditional method for L-tryptophan concentration detecting is time consuming.The calibration model was established based on the near infrared spectroscopy and the method of partial least square.In order to examine the stability and accuracy of the model,we compared the predicted value with the accurate value of L-tryptophan concentration,and the average relative error was 5.16%.Conclusion:The model has good prediction ability in providing reliable data for L-tryptophan concentration rapid detection in Escherichia coli fermentation process,and provide theoretical and practical basis for L-tryptophan fermentation process control.
