摘 要:以平菇菌丝体为材料,采用直接法、甲苯法及乙醇-氯仿法进行SOD提取,检测酶活性。结果表明,3种提取方法均能获得SOD,通过比较确定乙醇-氯仿法提取效果最佳。提取条件为:1 g平菇菌丝体加入8 ml 0.2 mol/L的Na HCO3溶液、6 ml乙醇-氯仿(体积比5/3)溶液,搅拌2 h,4 000 r/min离心30 min。此法提取得到的粗酶液比活力最高,为1.345 U/mg;纯化后的比活力为1.521 U/mg;纯化倍数为1.720;回收率为91.7%。SOD was extracted from Pleurotus ostreatus mycelium with direct method, toluene method and ethanol-chloroform method respectively, and the enzyme activity was tested. The results showed that all the three extraction methods can obtain SOD, and the extraction effect of ethanol - chloroform method was the best through the comparison. Extraction conditions was adding 1 g mushroom mycelium into 8ml NaHCO3 solution of 0.2 mol/L and 6ml ethanol - chloroform solution, then centrifuging 30min at 4 000 r/min after stirring 2 h. The crude enzyme activity of 1.34515 U/p~ g was the highest. After purifying the specific activity was 1.521 U/mg. The purification multiple was 1.720, and the recovery rate was 91.7%.
