摘  要:建立高效液相色谱法同时测定玉米蛋白粉中17种游离氨基酸含量的方法。样品用盐酸溶液(0.1mol/L)提取,采用6-氨基喹啉-N-羟基琥珀酰亚胺基-氨基甲酸酯(AQC)为衍生剂衍生。应用No—va—PakTM C18柱,以10mmoL/L醋酸铵缓冲液、乙腈和超纯水为流动相进行梯度洗脱,柱温为37oC,检测波长为248nm,外标法定量。结果表明,17种游离氨基酸线性关系良好,其中16种游离氨基酸在0.0025~0.5μmoL/mL范围内具有良好的线性关系,胱氨酸浓度在0.0063~1.25μmol/mL范围内线性关系良好,相关系数在0.9991~0.9999之间,回收率为95.10%~105.21%,RSD为1.36%~4.39%。该方法灵敏度高、重现性好、操作简单,可用于检测玉米蛋白粉中游离氨基酸含量。High performance liquid chromatography was to be established along with a method for the determina- tion of 17 kinds of free amino acids in corn gluten meal by high performance liquid chromatography. The specimen was extracted with hydrochloric acid solution (0.1 mol/L) , and 6 -ami -noquinolyl -N -hydroxysuccinimidyl car- bamate (AQC) then was derivatized from the derivating agent. Gradient elution was achieved by using a Nova - Pak- TM C 18, and ammonium acetate buffer solution( 10 mmol/L) , acetonitrile and deionized water as the mobile phase with the column temperature of 37 ℃, the detected wave length of 248 nm. The external standard method was used for quantification. Results show there was a good linear relationship for 17 free amino acids, of which a good linear relationship was identified in 16 free amino acids in the range of 0. 002 5 - 0.5 μmol/mL, while cystine in the range of 0. 006 3- 1.25 μmoL/ml. The correlation coefficient is in the range of 0. 999 1 -0. 999 9. Average recovery of 17 amino acids was 95.10% - 105.21% , and the RSD was in the range of 1.36%-4.39%. This method is highly sen- sitive, reproducible , simple and suitable for the determination of free amino acids in corn gluten meal.
