摘  要:目的探讨甘草次酸(GA)对人胃癌SGC7901细胞增殖的影响及其可能机制。方法常规培养人胃癌SGC7901细胞,分为实验组1~7组和阴性对照组,实验组1~7组为培养SGC7901细胞加入GA的二甲基亚砜(DMSO)溶液,使GA的终浓度分别为6.25、12.50、25.00、50.00、100.00、150.00、200.00μmol·L^(-1),阴性对照组加入和终浓度GA相同体积的DMSO。GA作用48 h,采用四甲基偶唑蓝(MTT)法检测GA对人胃癌SGC7901细胞的增殖作用,用流式细胞仪检测碘化丙啶单染细胞的DNA含量。结果 MTT法测定结果显示,终浓度为6.25、12.50、25.00、50.00、100.00、150.00、200.00μmol·L^(-1)的GA对人胃癌SGC7901细胞抑制率分别为(8.27±1.80)%、(13.74±3.16)%、(25.85±4.35)%、(46.03±5.88)%、(97.78±1.10)%、(99.56±0.21)%、(98.86±0.35)%;GA终浓度低于100.00μmol·L^(-1)时,人胃癌SGC7901细胞抑制率随GA浓度的增加而逐渐增加,呈明显的剂量依赖关系(P<0.05)。流式细胞仪检测结果显示,终浓度为6.25、12.50、25.00、50.00、100.00、150.00、200.00μmol·L^(-1)的GA作用人胃癌SGC7901细胞48 h后,DNA处于亚二倍体、二倍体、四倍体、介于二倍体和四倍体之间的细胞数量差异有统计学意义(P<0.001);浓度低于150.00μmol·L^(-1)时,亚二倍体的细胞数量逐渐增加,且呈浓度依赖性(P<0.05);浓度高于150.00μmol·L^(-1)后,亚二倍体的细胞数量逐渐减少;浓度低于100.00μmol·L^(-1)时,二倍体的细胞数量逐渐增加,介于二倍体与四倍体之间、四倍体的细胞数量则逐渐减少;当浓度超过100.00μmol·L^(-1)时,二倍体的细胞数量逐渐减少,四倍体的细胞数量逐渐增加,介于二倍体与四倍体之间的细胞数量变化不大。结论 GA对人胃癌SGC7901细胞的生长有抑制作用,其抑制过程可能有细胞凋亡的参与,也有周期的改变。Objective To explore the effect of glycyrrhetinic acid( GA) on proliferation of human gastric cancer SGC7901 cells and the possible mechanism. Methods The human gastric cancer SGC7901 cells were cultivated routinely,and divided into the experimental 1- 7 groups and the negative control group. The cells of the experimental 1- 7 groups were cultured in dimethylsulfoxide( DMSO) with every final concentration of GA that was 6. 25,12. 50,25. 00,50. 00,100. 00,150. 00 and 200. 00 μmol·L^(-1). The cells of the negative control group were cultured in the volume of dimethylsulfoxide equaled to that of the GA end concentration. Cultured for 48 h,cells in each group were checked by methyl thiazolyl tetrazolium( MTT)method to determine the effect of GA on proliferation of gastric cancer SGC7901 cells. The flow cytometry was used to detected the content of DNA in the cells which were stained with propidium iodide. Results MTT results showed that the inhibition ratio of 6. 25,12. 50,25. 00,50. 00,100. 00,150. 00 and 200. 00 μmol·L^(-1)GA on human gastric cancer SGC7901 cells was( 8. 27 ± 1. 80) %,( 13. 74 ± 3. 16) %,( 25. 85 ± 4. 35) %,( 46. 03 ± 5. 88) %,( 97. 78 ± 1. 10) %,( 99. 56 ± 0. 21) % and( 98. 86 ± 0. 35) % respectively. When the concentration of GA was less than 100. 00 μmol·L^(-1),the growth inhibition ratio of human gastric cancer SGC7901 cells increased with the increasing of concentration of GA( P 0. 05). Flow cytometry results showed that there was statistic difference of the cell count which had hypodiploid DNA,diploid DNA( 2N),tetraploid DNA( 4N) and DNA between diploid and tetraploid( 2N- 4N)( P 0. 001) when 6. 25,12. 50,25. 00,50. 00,100. 00,150. 00 and 200. 00 μmol · L- 1GA acted to gastric cancer SGC7901 cells for 48 h.
