摘  要:目的:研究香菇多糖(LTN)对溃疡性结肠炎(UC)大鼠的改善作用及其机制。方法:将64只SD大鼠随机分为正常组(12只)和造模组(52只)。造模组大鼠采用2,4-二硝基氯苯丙酮和2,4-二硝基氯苯乙醇诱导UC模型。将造模成功的大鼠随机分为模型组、阳性组(柳氮磺胺吡啶片,300mg/kg)和LTN低、中、高剂量组(0.2、0.4、0.8mg/kg),每组10只。各给药组大鼠ig相应药物,正常组和模型组大鼠ig生理盐水,每天1次,连续15 d。检测外周血淋巴细胞(PBMC)中CD_3^+、CD_4^+、CD_8^+水平和结肠组织中肿瘤坏死因子α(TNF-α)、白细胞介素2(IL-2)、IL-4、IL-10水平,并观察结肠组织病理变化。结果:与正常组比较,模型组大鼠PBMC中CD_3^+、CD_4^+、CD_8^+水平和CD_4^+/CD_8^+比值以及结肠组织中IL-4、IL-10水平明显降低,结肠组织中TNF-α、IL-2水平明显升高(P<0.05),结肠组织发生明显溃疡性病变(集中于Ⅱ、Ⅲ级)。与模型组比较,各给药组大鼠PBMC中CD_3^+、CD_4^+、CD_8^+水平,CD_4^+/CD_8^+比值以及结肠组织中IL-4、IL-10水平明显升高,结肠组织中TNF-α、IL-2水平明显降低(P<0.05);结肠组织炎症明显减轻、溃疡形成明显改善。结论:香菇多糖对UC模型大鼠具有一定的改善作用,其机制可能与增强大鼠免疫功能有关。OBJECTIVE:To study the regulatory effect of lentinan(LTN) on immune function in ulcerative colitis(UC) rats and its mechanism.METHODS:64 SD rats were randomly divided into normal group(12 rats) and modeling group(52 rats).UC model was induced in modeling group by a compound method of 2,4-dinitrochlorobenzene(DNCB) acetone and 2,4-DNCB ethanol.After modeling,rats were randomly divided into model group,positive group(sulfasalazine tablet,300 mg/kg),LTN low-dose,medium-dose and high-dose groups(0.2,0.4,0.8 mg/kg),with 10 rats in each group.Treatment groups were given relevant medicine intragastrically,and normal group and model group were given normal saline intragastrically,once a day,for consecutive 15 d.The levels of CD3+,CD4+,CD8+ in peripheral blood lymphocytes(PBMC) and the levels of TNF-α,IL-2,IL-4 and IL-10 in colon were all determined,and the pathological changes of colon tissue were observed.RESULTS:Compared with normal group,the levels of CD3+,CD4+,CD8+,the ratio of CD4/CD8+,and the levels of IL-4 and IL-10 decreased significantly,while the levels of TNF-α and IL-2 in colon tissue significantly increased(P〈0.05);obvious ulcerative lesion was observed in colon tissue(concentrated in grade Ⅱ and Ⅲ).Compared with model group,the levels of CD3+,CD4+,CD8+,the ratio of CD4/CD8+,and the levels of IL-4,IL-10 increased significantly,while the levels of TNF-α,IL-2 in colon tissue significantly decreased(P〈0.05);the inflammation of colon tissue was significantly reduced,and the ulcer formation was significantly improved.CONCLUSIONS:LTN has a certain improvement effect on UC model rats,and its mechanism may be related to enhancing the immune function.
