摘 要:目的探讨黑木耳多糖对小鼠营养性肥胖干预作用及可能机制。方法 40只小鼠随机分为对照组、高脂组、黑木耳多糖低、中、高剂量组,高脂膳食同时灌胃给予黑木耳多糖,连续4周;测定各组小鼠体重、Lee's指数、脂肪指数、血清中总胆固醇(TC)、总甘油三酯(TG)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)、瘦素及激素敏感性脂肪酶(HSL)含量。结果与对照组比较,高脂组小鼠Lee's指数、脂肪指数[分别为(341.53±4.39)、(3.59±0.83)%]明显升高,血清中TC、TG、瘦素、HSL含量[分别为(5.06±0.10)、(2.12±0.20)mmol/L、(4.00±0.25)、(8.16±0.91)ng/m L]明显升高;与高脂组比较,黑木耳多糖高剂量组(100 mg/kg)小鼠Lee's指数、脂肪指数[(328.48±4.16)、(2.37±0.14)%]下降,血清中TC、TG、瘦素含量[(3.50±0.21)、(1.71±0.17)mmol/L、(3.72±0.55)ng/m L]下降,黑木耳多糖中剂量组(50 mg/kg)小鼠血清中HSL含量[(7.79±0.25)ng/m L]下降。结论黑木耳多糖对小鼠高脂膳食所致营养性肥胖具有干预作用,其机制可能与降低小鼠血清中瘦素及激素敏感性脂肪酶水平有关。Objective To explore intervention effect and its possible mechanism of Auricularia polysaccharide on nutritional obesity in mice. Methods Forty mice were randomly divided equally into a control( with normal feed),high-fat( with high-fat feed),and low,medium and high dose Auricularia polysaccharide groups( with high-fat feed and intragastric administration of Auricularia polysaccharide). Four weeks after the continuous treatment,the body weight,Lee's index,fat coefficient,serum total cholesterol( TC),total triglyceride( TG),high density lipoprotein( HDL),lowdensity lipoprotein( LDL),leptin( LEP),and hormone sensitive lipase( HSL) of all the mice were determined. Results Compared with those of the mice in the control group,Lee 's index( 341. 53 ± 4. 39),fat index( 3. 59 ± 0. 83%),TC( 5. 06 ± 0. 10 mmol/L),TG( 2. 12 ± 0. 20 mmol/L),LEP( 4. 00 ± 0. 25 ng /m L),and HSL( 8. 16 ± 0. 91 ng /m L)increased significantly in the mice of high-fat group. Compared w ith those of the high-fat group,Lee's index( 328. 48 ±4. 16),fat index( 2. 37 ± 0. 14%),TC( 3. 50 ± 0. 21 mmol / L),TG( 1. 71 ± 0. 17 mmol / L),and LEP( 3. 72 ± 0. 55 ng / m L) decreased significantly in the mice treated w ith high dose Auricularia polysaccharide( 100 mg / kg) and serum LEP( 7. 79 ± 0. 25 ng / m L) decreased significantly in the mice treated w ith medium dose Auricularia polysaccharide( 50 mg /kg). Conclusion Auricularia polysaccharide has antagonistic effect on nutritional obesity and its mechanism may be related to decreased LEP and HSL levels in mice.
