摘  要:目的探讨制备抑制翼状胬肉术后血管增生免疫纳米微球,即血管内皮生长因子单克隆抗体.聚乳酸(汉防已甲素)。纳米微粒[VEGFMcAb—PLA(Tet)-NP]的制备方法。方法采用复乳法,以天然中药Tet作为试验药物,以PLA为包封材料,制备汉防已甲素聚乳酸纳米微粒·PLA(Tet)-NP]。观察其表面形态、结构、粒径及电动电位(Zeta电位)。采用碳二亚胺法将Tet聚乳酸纳米微球PLA(Tet)-NP与抗人VEGFMcAb连接,制备免疫纳米微球。观察其表面形态、结构、粒径及Zeta电位。结果成功制备出了PLA(Tet)-NP及VEGFMcAb—PLA(Tet)-NP。微粒的表征:类圆形囊泡,均质,粒径大小约为200-242nm,Zeta电位为-7.7my。结论通过聚乳酸为包封材料,可成功制备出载药PLA(Tet)-NP,采用碳二亚胺法与抗人VEGFMcAb成功连接。为今后进一步开展抑制翼状胬肉术后血管增生的研究奠定了基础。Objective Preparation of tetrandrine (Tet) polylactic acid immunonanoparticles (INP) nanoparticles, as a basis for study of vascular proliferation after pterygium surgery. Methods Using double emulsion method, with natural Chinese herbal medicine Tet as experimental drug, using polylactic acid (PLA) as embedding materials, preparation of Tet polylactic acid nanoparticles-PLA (Tet)-NP. To observe the surface morphology, structure, particle size and Zeta potential. Anti-VEGF McAb was attched to PLA (Tet)-NP by Carbodiimide method, preparation of Tet polylactic acid immunonanoparticles nanoparticles- VEGF McAb-PLA(Tet)-NP. To observe the surface morphology, structure, particle size and Zeta potential. Results PLA (Tet)-NP and VEGF McAb-PLA (Tet)-NP were prepared successfully, particle characterization : round vesicles, homogeneous, the particle size is about 200-242nm, the Zeta potential of - 7.7my. Conclusion The polylactic acid as embedding materials, can be successfully prepared drug loaded PLA (Tet) -NP, to investigate the feasibility of the nanoparticles as drug carrier. VEGF McAb-PLA (Tet)- NP can be successfully prepared by Carbodiimide method. For further inhibit angiogenesis after pterygium surgery research foundation.
