摘 要:目的:研究分析人类runt相关转录因子3(RUNX3)蛋白在肝癌组织中的表达变化及其临床意义。方法采用免疫组化法对84例肝癌组织标本(肝癌组)及40例癌旁组织标本(癌旁组)进行染色观察,比较RUNX3蛋白在2组标本中的表达变化,并分析RUNX3蛋白在肝癌发生、发展中的作用。结果肝癌组织中RUNX3蛋白阳性表达率高于癌旁肝组织(χ^2=14.849,P<0.05)。Ⅰ+Ⅱ期肝癌患者的RUNX3蛋白阳性表达率高于Ⅲ+Ⅳ期患者(χ^2=5.601,P<0.05);高、中分化患者的RUNX3蛋白阳性表达率为75.56%显著高于低分化患者的17.95%(χ^2=8.697,P<0.05);未发生淋巴结转移患者的RUNX3蛋白阳性表达率高于发生淋巴结转移患者(χ^2=7.571,P<0.05);而不同年龄、性别、肿瘤部位、组织学类型、肿瘤大小、是否存在包膜、是否存在门脉癌栓患者RUNX3蛋白阳性表达率差异无统计学意义(P>0.05)。结论肝癌组织中RUNX3蛋白表达较癌旁组织下调,并且与肝癌的分期、分化程度、淋巴结转移有关,RUNX3蛋白下调表达与肝癌的发生、发展具有一定的关系。Objective To investigate the level of human runt -related transcription factor 3 ( RUNX3 ) protein in hepatocellular carcinoma and its clinical significance .Methods A total of 84 pathologically diagnosed samples of hepa-tocellular carcinoma (an HCC group) and 40 samples of adjacent tissues (an adjacent group) were selected.Then, im-munohistochemical staining was used to observe the levels of RUNX 3 in both groups .Meanwhile , the role of RUNX3 in the development of hepatocellular carcinoma was analyzed .Results Among 80 HCC samples were 41 (48.81%) posi-tive RUNX3 cases, in contrast with 34 (85.0%) positive RUNX3 cases in 40 samples of adjacent tissues , indicating sta-tistical difference between both groups (χ^2 =14.849,P〈0.05).HCC patients at Stages Ⅰand Ⅱ produced a positive RUNX3 protein rate of 58.18%, which was significantly higher than 31.03% from patients at Stages Ⅲ+Ⅳ (χ^2 =5.601,P〈0.05).The positive RUNX3 protein rate was 75.56%in high and medium differentiated patients , which was remarkably higher than 17.95%in patients with low differentiation (χ^2 =8.697,P〈0.05).The positive RUNX3 pro-tein rate was 56.92%in patients without lymph node metastasis , which was remarkably higher than 21.05%in patients with lymph node metastasis (χ^2 =7.571,P〈0.05).No significant difference was found between both groups as to age , gender, tumor site, histological type, tumor size, the presence of capsule, the possibility of changes in a positive RUNX3 protein rate for patients with portal vein tumor thrombus (P〉0.05).Conclusion The level of RUNX3 protein in HCC tissues is lower than that in the adjacent tissues , which is related to the pathological stage , differentiation degree and lymph node metastasis of HCC .Down-regulation of RUNX3 protein is associated with the development of HCC .
