摘 要:【目的】评估从江香猪CYP2A19基因的纯合度及遗传变异,为开发从江香猪药物代谢模型积累基础资料。【方法】应用SNa Pshot检测方法对141份从江香猪样本CYP2A19基因编码区rs323914689、rs81217981、rs333289891、rs343272409位点及3'端非翻译区rs338584662和rs321736682位点进行分型分析,并计算从江香猪种群的基因型频率、等位基因频率、有效等位基因数(Ne)、杂合度(He)、多态位点数及多态位点百分比。【结果】除rs333289891位点表现为单态外,其余5个位点均检测到2种或3种基因型,多态位点百分比为83.33%。χ2检验结果显示,5个多态位点均处于Hardy-Weinberg平衡状态(P〉0.05)。5个多态位点的Ne介于1.0071~1.3129,He介于0.0071~0.2057。【结论】不同品系猪种由于遗传背景差异而具有不同的单核苷酸变异,其药物代谢特性也因此存在差异。从江香猪群体纯合度高、遗传稳定性好、变异程度低,是其作为药物代谢动物模型的关键基础。[ Objective ]Homozygositiy and genetic variations of CYP2A19 gene in Congjiang Xiang-pig were assessed, in order to accumulate basic informations for developing drug metabolism model. [Method]A total of 141 Congjiang Xiang-pig samples were collected from Congjiang county, Guizhou. Genotypes of loci rs323914689, rs81217981, rs333289891 and rs343272409 in CYP2AI9 gene coding region and rs338584662 and rs321736682 loci in 3'-end non-translation regions were detected by SNaPshot method. Then genotype frequencies, allele frequencies, numbers of effective alleles (Ne), heterozygosities (He), number and percentage of polymorphie loci of Congjiang Xiang-pig population were calculated. [Result]The results showed that, only rs333289891 locus was monomorphie, other 5 loci were polymorphie, with 2 or 3 genotypes, and the percentage of polymorphic loci was 83.33%. Chi-square test results showed that, all 5 polymorphic loci were in the Hardy-Weinberg equilibrium state (P〉0.05). In 5 polymorphic loci, Ne ranged from 1.0071 to 1.3129, and He ranged from 0.0071 to 0.2057. [Conclusion]Due to difference in genetic background, different breeds of pigs have different mononueleotide variations, and their drug metabolism characteristics are also different. Cangjiang Xiang-pig can taken as animal model of drug metabolism because it has characteristics of high homozygosity, good genetic stability and low variation.
