摘  要:目的:观察姜黄素(curcumin)对APPswe/PS1d E9双转基因小鼠认知功能的影响以及对海马组织中α-突触素(α-synuclein)和微管相关蛋白2(microtubule associated protein-2,MAP2)表达的影响,并探讨其可能的机制。方法:将3月龄APPswe/PS1d E9双转基因小鼠随机分为未处理组、姜黄素低剂量组和姜黄素高剂量组,每组10只,未处理组给予正常饲料,低、高剂量姜黄素组按照160、1 000 mg/kg拌饲喂养,连续喂养6个月,采用Morris水迷宫检测姜黄素对双转基因鼠空间学习记忆能力的影响;采用免疫组化检测姜黄素对双转基因鼠海马中α-synuclein和MAP2表达的影响。结果:Morris水迷宫实验表明姜黄素可以明显改善双转基因鼠学习记忆能力(P<0.05);免疫组化结果显示与未处理组相比,低、高剂量姜黄素处理组小鼠海马内α-synuclein阳性细胞数均减少(P<0.05),呈剂量依赖性;但MAP2阳性表达随着姜黄素浓度的升高而增加(P﹤0.05)。结论:姜黄素可能通过抑制α-synuclein表达并增强MAP2的表达,从而改善AD转基因鼠的认知和记忆功能起到神经保护作用。Objective:To observe the effects of curcumin on cognitive function of APPswe/ PS1 d E9 double transgenic mice and the expression of α-synuclein and MAP2 in hippocampus,and to explore the potential mechanisms. Methods:The 3-month-old APPswe/PS1 d E9 double transgenic mice were randomly divided into untreated group(n=10),curcumin low-dose group(n=10)and curcumin high dose group(n=10). Normal diet,diet mixed with 160 mg/kg curcumin and diet mixed with 1 000 mg/kg curcumin were given,respectively. The Morris water maze test was used to detect the effects of curcumin on spatial learning and memory of AD mice. IHC was carried out to evaluate the expression of α-synuclein and microtubule associated protein-2(MAP2)in hippocampus. Results:Morris water maze test showed that curcumin could improve learning and memory of AD mice. IHC results showed that compared with those of untreated group,the expressions of α-synuclein in the hippocampus were significantly reduced in low and high dose of curcumin group;however,the expressions of MAP2 were significantly increased in curcumin treatment group in a dose-dependent manner(P〈0.05). Conclusion:Curcumin could play a role in improving cognition and memory ability of AD transgenic mice via upregulating the expression of MAP2 and down-regulating α-synuclein.
