摘 要:通过自水解预处理板栗栗苞,以预水解液组成增殖培养基培养米根霉,增殖的米根霉再利用栗苞酶解液生产富马酸。结果表明:220℃自水解预处理栗苞,有效疏解栗苞紧密的木质纤维结构,以50 FPIU(以1 g纤维素计)纤维素酶水解50 g/L预处理栗苞,酶解得率大于95%;经增殖培养基培养米根霉,菌体生物量达4.5 g/L;增殖的米根霉利用栗苞酶解液发酵产富马酸,富马酸质量浓度为15.78 g/L,糖酸转化率为0.34 g/g。通过两段式发酵工艺,米根霉有效利用板栗栗苞生产富马酸。Fumaric acid production from autohydrolysis?pretreated Chinese chestnut shell by Rhizopus oryzae was investigated in a two?stage fermentation. Autohydrolysis pretreatment of Chinese chestnut shell was carried out at 220 ℃. Enzymatic hydrolysis yield of pretreated Chinese chestnut shell was over 95%based on 50 g/L substrate loading using a cellulase loading of 50 FPIU/g cellulose. Rhizopus oryzae could grow and obtain 4. 5 g/L fungal biomass in proliferation medium containing xylose.Pre?cultured Rhizopus oryzae was used for production of fumaric acid from enzymatic hydrolysate of pretreated Chinese chestnut shell. Fumaric acid of 15. 78 g/L was obtained after 96 h fermentation with a yield of 0. 34 g/g.
