摘  要:α-淀粉酶(Amy)是一类关键的淀粉水解酶,在香蕉果实成熟淀粉降解转化为可溶性糖的过程中发挥着重要作用。本研究基于香蕉基因组数据,通过生物信息学的方法对香蕉α-淀粉酶基因的基本理化性质、二级结构预测、亚细胞定位、内含子和外显子结构和保守结构域进行初步的预测与分析,并构建系统发育树。结果表明,编码香蕉α-淀粉酶的基因有12个,根据在染色体的位置命名为MaAmy1~12,MaAmy基因家族编码的氨基酸的范围在79~914个,编码的蛋白相对分子量介于8.8~103.2kD之间,12个MaAmy蛋白中有8个偏酸性,3个偏碱性。不稳定指数分析发现,8个MaAmy蛋白为稳定蛋白;疏水性分析表明,所有的MaAmy蛋白为亲水性蛋白;香蕉MaAmy家族最多的含有13个内含子;进化分析表明,MaAmy家族分4个亚家族。α-amylase is a key starch-degrading enzyme,which plays an important role in the process of starch degradation into soluble sugar during banana fruit ripening.The basic physical and chemical characters,secondary structure,subcellular location,gene structure,evolutionary relationship and conserved motif of bananaα-amylase gene were analyzed and phylogenetic trees were constructed by bioinformatics methods.The results showed that there were 12 genes codingα-amylase,which were named from MaAmy1 to MaAmy12based on their chromosomal location.The identified banana Amygene family members encode predicted proteins ranging from 79 to 914amino acid residues(aa)in length,and computed molecular masses of 8.8kD to 103.2kD.There were 8acidic proteins and 3alkalinity proteins in the MaAmy family.Instability index analysis found that 8of the MaAmy protein was stabilized protein.Hydrophobic analysis showed that all of the MaAmy proteins are hydrophilic proteins.MaAmy gene family mostly contained thirteen introns.Evolution analysis indicated that MaAmy family was classed into two subfamilies.
