摘  要:为了克隆鹅细小病毒(GPV)NS1基因,并对其进行生物信息学分析,试验参照GenBank发表的GPV全基因组序列(U25749)设计扩增GPV-NS1基因的1对特异引物,经PCR扩增,将克隆的GPV-NS1基因进行TA克隆和转化,对鉴定正确的质粒进行测序,并对测序结果进行生物信息学分析。结果表明:YBLJ株GPV-NS1基因全长1884bp,编码627个氨基酸,与其他11株GPV的NS1基因核苷酸同源性为93.6%~99.9%,氨基酸同源性为96.2%~99.8%;蛋白二级结构中α螺旋占31.42%,β折叠占19.30%,无规则卷曲占49.28%;蛋白三级结构中α螺旋和β折叠占GPV-NS1总蛋白的一半以上的比例。说明GPV-NS1基因和其编码的氨基酸均较保守,尤其氨基酸更为保守,提示NS1蛋白虽然是非结构蛋白,但其对于GPV应该是至关重要的功能蛋白。To clone the NS1 gene of goose parvovirus (GPV) and conduct a bioinformatics analysis of the NS1 gene, a pair of specific primers was designed according to the published whole sequence of GPV(U25749) in GenBank, and then the GPV - NS1 gene was amplified by PCR. The cloned GPV - NSI gene was used for TA cloning and transformation, the verified correct plasmid was sequenced, and the sequenced result was used for bioinformatics analysis. The results showed that the full length of GPV - NS1 gene of YBLJ strain was 1 884 bp, encoding a protein of 627 amino acids. The GPV - NS1 gene of YBLJ strain shared 93.6% to 99.9% nueleotide homology and 96.2% to 99.8% amino - acid homology with those of other eleven strains of goose parvovirus. The alpha helixes, beta sheets, and random coils accounted for 31.42% , 19.30% ,49.28% in the secondary structure of GPV- NS1 protein, respective/y, and the alpha helixes and beta sheets accounted for more than half of the proportion of GPV - NS1 total protein in the tertiary structure of protein. The results indicate that GPV - NS1 gene and its enco- ded amino acids are conservative, espeeially the amino acids are more conservative. It is suggested that GPV - NS1 protein is a non - structural protein, but it should be an essential functional protein for GPV.
