乙酰葛根素对Aβ_(25-35)诱导BV-2小鼠小胶质细胞株caspase-8和caspase-3表达的影响

摘　　要：目的观察乙酰葛根素对β淀粉样蛋白(Aβ25-35)诱导的BV-2小鼠小胶质细胞株caspase-8和caspase-3表达的影响。方法将细胞分为对照组、模型组(采用凝聚态Aβ25-35刺激BV-2小鼠小胶质细胞株建立细胞模型)、乙酰葛根素低、中、高剂量干预组及caspase-8抑制剂(Z-IETD-fmk)组共6组。用Western blot检测caspase-8、caspase-3蛋白表达;实时荧光定量PCR(real-time PCR)检测caspase-8、caspase-3 mRNA表达。结果与对照组相比,模型组caspase-8、caspase-3蛋白表达明显上调(P<0.01),caspase-8、caspase-3基因表达明显增加(P<0.01);与模型组相比,乙酰葛根素各剂量组均可不同程度地明显下调caspase-8、caspase-3蛋白表达(P<0.01),明显抑制caspase-8、caspase-3基因表达(P<0.01)。结论乙酰葛根素可剂量依赖性地抑制Aβ25-35诱导BV-2小鼠小胶质细胞株caspase-8、caspase-3的表达。Objective To investigate the effect of acetylpuerarin on expression of caspase-8 and caspase-3 in BV-2 mouse microglia cell line induced by Aβ25-35. Methods BV-2 microglia cells were divided into six groups：control group,model group（ cells were activated with condensed Aβ25-35 as cell model）,acetylpuerarin intervention groups（ low-dose group,moderate-dose group,high-dose group） and caspase-3 inhibitor（ Z-IETD-fmk）group. The protein expression of caspase-8 and caspase-3 was detected by Western blot. The mRNA expression of caspase-8 and caspase-3 was measured by real-time PCR. Results Compared with control group,protein expression of caspase-8 and caspase-3 in model group was significantly up-regulated（ P〈 0. 01）,and gene expression of caspase-8 and caspase-3 markedly increased（ P〈 0. 01）. Compared with model group,protein expression of caspase-8 and caspase-3 in acetylpuerarin each dose group was significantly down-regulated（ P〈 0. 01）,and gene expression of caspase-8 and caspase-3 was markedly decreased（ P〈 0. 01）. Conclusions Acetylpuerarincan inhibit caspase-8 and caspase-3 expression of BV-2 mouse microglia cell line induced by Aβ25-35 in a dosedependent manner.
