摘  要:目的:观察茶黄素对大鼠缺血性脑损伤所致炎症反应的作用。方法:64只健康成年SD大鼠随机分为4组:假手术组、模型组与茶黄素低(20 mg·kg-1)、高(40 mg·kg-1)剂量组。连续给药7 d后,除假手术组外,采用大脑中动脉线栓法(MCAO)制备大鼠实验性脑缺血模型。缺血24 h后,进行神经功能评分,然后每组随机抽取8只计算脑组织含水量,另外8只用于测定脑组织核因子-κB p65(NF-κB p65)mRNA水平。最后,测定血清中肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)及细胞间黏附分子-1(ICAM-1)含量。结果 :与假手术组相比,模型组大鼠神经功能评分、脑组织含水量、脑组织NF-κB p65 mRNA表达量、血清TNF-α、IL-1β及ICAM-1含量显著升高。与模型组相比,茶黄素高剂量组脑组织NF-κB p65 mRNA表达量明显减少,茶黄素低、高剂量组大鼠神经功能评分、脑组织含水量、血清TNF-α、IL-1β及ICAM-1含量显著降低。结论:茶黄素对大鼠缺血性脑损伤造成的炎症反应有一定的缓解作用。OBJECTIVE To investigate effects of theaflavin on inflammation in rats caused by ischemic brain injuries.METHODS Sixty four SD rats were randomly divided into 4 groups:sham operation group(Sham),model group(Model),theaflavin low(20 mg·kg-1)and high(40 mg·kg-1)dose groups.Seven days after continuous administration,in addition to Sham group,ischemic brain injuries in rats were induced by middle cerebral artery occlusion(MCAO).After that for 24 h,neurological score was calculated for rats.Then water content of brain tissues in rats was calculated in 8 rats in each group,expression of NF-κB p65 mRNA was determined in another 8 rats.Finally,TNF-α,IL-1βand ICAM-1 contents of serum were detected in each group.RESULTS Compared with Sham group,neurological score,water content of brain tissues,expressions of NF-κB p65 mRNA,TNF-α,IL-1βand ICAM-1levels in model group were elevated significantly.Compared with Model group,expression of NF-κB p65 mRNA in brain tissues was decreased obviously in theaflavin high dose group.Moreover,neurological score,water content of brain tissues,TNF-α,IL-1βand ICAM-1 levels were reduced significantly in theaflavin low and high dose groups.CONCLUSION Theaflavin can alleviate inflammation caused by ischemic brain injuries in rats to a certain degree.
