摘 要:采用稀释涂布法分离了浓香型白酒窖池窖泥中的好氧细菌,并利用形态学特征结合16S rRNA基因序列分析技术对分离的细菌进行鉴定,探讨窖泥可培养好氧细菌的群落结构。结果表明,根据菌落和细胞形态特征,分离到了1株无芽孢短杆菌和18株芽孢杆菌,均为革兰氏阳性;采用16S rRNA基因序列分析,将19株细菌鉴定为9个种:米氏解硫胺素芽孢杆菌、枯草芽孢杆菌、菠茨坦短芽孢杆菌、地衣芽孢杆菌、侧孢短芽孢杆菌、短小芽孢杆菌、Paenibacilluslautus、耐寒短杆菌和P.cellulositrophicus;其中波茨坦短芽孢杆菌、米氏解硫胺素芽孢杆菌和P.lautus鲜有浓香型白酒窖泥中分布的报道。本研究揭示了浓香型白酒窖泥可培养好氧细菌的群落结构,为改善窖泥质量、提高白酒品质等提供了理论依据。s:In the experiment, aerobic bacteria in pit mud of Nongxiang Baijiu were isolated by dilution plating method, and then identified by morphological characteristics coupled with sequence analysis of 16S rRNA gene. The results indicated that, one strain of non spore-producing short rod-shaped bacterium and 18 strains of spore producing Gram-positive bacteria were isolated, and those 19 strains were identified as 9 species by 16S rRNA gene sequencing analysis, including Aneurinibacillus migulanus, Bacillus subtilis, Brevibacillus borstelensis, Ba. licheni-formis, Br. laterosporus, Ba. pumilus, Paenibacillus lautus, Brevibacterium frigoritolerans and P. cellulositrophicus. Among them, the distribu-tion of Br. Borstelensis, A. migulanus and P. Lautus in pit mud of Nongxiang Baijiu were seldom reported. This study revealed the community structure of aerobic bacteria in pit mud of Nongxiang Baijiu, and provided theoretical evidence for improving pit mud quality and improving li-quor quality.
