摘  要:目的本文利用tau基因敲除小鼠模型,观察tau基因表达水平的变化对中脑部位Nurr1蛋白表达的影响。方法取同窝出生的同性别的野生型(Tau+/+)、杂合子(Tau+/-)、纯合子(Tau-/-)系列tau基因敲除小鼠。应用免疫印迹技术定量检测不同年龄3种基因型小鼠中脑部位Nurr1的表达水平;应用免疫荧光染色的方法,定位定量检测18月龄3种基因型小鼠中脑黑质致密部(SNC)和腹侧背盖区(VTA)Nurr1和TH的表达水平。结果免疫印迹的结果显示Nurr1在6月龄至24月龄Tau-/-小鼠中脑的表达量增多,与Tau+/+和Tau+/-小鼠相比差异有统计学意义(P<0.05);在免疫荧光实验中,Nurr1在Tau-/-小鼠中脑SNC和VTA部位TH阳性神经元中表达量增多,与Tau+/+和Tau+/-小鼠相比差异有统计学意义(P<0.05);3种基因型小鼠中脑部位TH表达量差异无统计学意义。结论 Nurr1在成年Tau基因敲除小鼠中脑多巴胺神经元中表达上调,而中脑TH表达水平无改变,Nurr1可能参与维持TH的表达。Object To anaylsis the expression of Nurrl in tan-knockout mice model midbrain under different levels of tau expression. Methods We conducted western bolt analysis to detect the expression of Nurrl in the midbrain of tan-knock mice at the age of 6, 12, 18 and 24 month, separately. (Including genotype Tan+/+, Tau+/- and Tan-/- ). We measured the expression of Nurrl and TH by using immunofluorescence technique in the SNC and VTA of tau-knock mice at 18-month-old. Results West- ern blot results showed that the expression of Nurrl in the midbrain of 6-, 12-, 18- and 24- month-old Tau-/- mice was significant- ly upregnlated (P〈0.05). A significantly increase of nuclear Nurrl expression was observed in TH+ neurons of 18-month-old Tan-/- mice compared with control Tan+/+ mice and Tan+/- mice (P〈0.05) in the SNC and VTA. However, there was no significant differ- ence in the expression of TH in the midbrain. Conclusion The expression of Nurrl in midbrain TH+ neurons of Tau-/- mice was significantly upregulated, while there was no variety in the expression of TH in the midbrain, which suggested that Nurrl may main- tain the expression of TH.
