乳岩宁方配合依西美坦对乳腺癌荷瘤裸鼠瘤重及GLUT1和PKM2表达的影响

摘　　要：目的:观察中药复方乳岩宁联合依西美坦对乳腺癌荷瘤裸鼠瘤重及GLUT1和PKM2蛋白表达的影响,探讨乳岩宁干预内分泌治疗的作用机制。方法:建立乳腺癌(MDA-MB-435)荷瘤裸鼠模型。将造模成功的裸鼠随机分为依西美坦组、乳岩宁组、联合组(依西美坦+乳岩宁组)及空白对照组。日1次灌胃,连续21天。将裸鼠断颈法处死,剥离肿瘤组织并测出瘤体重量。采用Western Blotting方法检测瘤组织中GLUT1和PKM2蛋白的表达。结果:(1)瘤重:乳岩宁、依西美坦和联合用药组瘤重均低于对照组,有统计学意义(P<0.05),其中依西美坦组的瘤重组低于乳岩宁组,并且结合组的瘤重最低,有显著性差异(P<0.05)。(2)GLUT1和PKM2蛋白的表达:对照组最高,联合组表达最低,依西美坦组低于乳岩宁组,实验结果有非常显著性差异(P<0.01)。结论:乳岩宁能提高依西美坦的抑瘤作用,可通过干预内分泌治疗来抑制肿瘤细胞的代谢,增强依西美坦的治疗效果。Objective： To observe the effect of Chinese herbal compound formula-Ruyanning combined with Exemestane on the tumor weight and the expressions of GLUT1 and PKM2 protein of tumor-bearing nude mice with breast cancer. To research the mechanisms of therapy of interfering endocrine with Ruyanning. Methods： The model of tumor-bearing nude mice with breast cancer（ MDA-MB-435） was established,and the modelled nude mouse were randomly divided into Exemestane group,Ruyanning group,joint group（ Ruyanning ＋ Exemestane） and control group. Then the nude mouse were administered the medicine by gavage,once a day,for 21 days. All the nude mouse were killed through breaking neck,and the tumor tissue was taken out completely and weighted. The expressions of GLUT1 and PKM2 protein in tumor tissue were detected by Western Blotting method. Results：Tumor weight： The tumor weights in Exemestane group,Ruyanning group and joint group was all lower than that in control group,which was statistically significant（P〈0. 05）. Among them,the tumor weight in joint group was lowest,and the tumor weight in Exemestane group was lower than that in Ruyanning group. There were significant differences（P〈0. 05）. The expressions of GLUT1 and PKM2 protein： The expressions of control group were highest,while the expressions of joint group were lowest. The expressions of Exemestane group were lower than those of Ruyanning group. There were very significant differences（P〈0. 01）. Conclusion： Ruyanning can improve the effect of Exemestane on tumor suppression,and inhibit the metabolism of tumor cell through therapy of interfering endocrine,thus enhance the therapeutic effect of Exemestane.
