摘 要:为探究大豆蛋白是否具有保肝作用,采用昆明种雄性小鼠作为试验对象,建立四氯化碳急性肝损伤模型,通过灌胃方式1次/d给予不同组别小鼠不同剂量、不同成分(大豆分离蛋白SPI、11S、7S)的蛋白(300、500、700 mg/kg),并设一组大豆蛋白饲料组(SPI占比为20%),持续15 d,于末次给药后注射CCl_4油溶液(10 mL/kg,0.15%)。通过测定血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)活力,肝脏中丙二醛(MDA)、谷胱甘肽(GSH)含量、谷胱甘肽过氧化物酶(GSH-Px)活力及肝脏指数评价保肝效果。结果显示,每天摄入700 mg/kg的SPI对由四氯化碳致肝损伤鼠有显著的保护作用;与模型组相比,700 mg/kg的7S蛋白能显著抑制由四氯化碳引起的血清中ALT、AST活力升高和小鼠肝脏中MDA含量的升高,并能显著提高其肝脏中的GSH含量、GSH-Px活力(P<0.05)。大豆蛋白可抵御CCl_4致化学性肝损伤,其中的7S蛋白是主要的保肝活性成分。The present study was designed to investigate the hepatoprotective effect of soybean protein isolate (SPI) against carbon tetrachloride -induced damage in KunMing male mice. SPI and 1 IS, 7S proteins at a dose of300, 500, 700 mg/kg was orally administered to mice, we also added a group of mice which was given SPI fodder (20% SPI was contained) for 15 days. On the last dose, 0.15% of CC14 (olive oil) were administered to mice fol- lowed last given drugs. The activity of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) in ser- um, malondialdehyde (MDA) and glutathione (GSH) level, glutathione peroxidase ( GSH - Px) activity in liver and liver index were determined. The result showed that pretreatment of mice with SPI (700 mg/kg) had significant hep- atoprotection against CC14 -induced liver damage. Compared with CC14 model group, 7S soy protein (700 mg,/kg) re- duced the activity of ALT, AST in serum and the MDA level in liver, and improved GSH level and GSH - Px activity significantly ( P 〈 0.05 ).
