摘  要:研究利用树脂对脂肪酶LipozymeCALB L进行固定化,然后将固定化酶用于催化富含二十二碳五烯酸(EPA)和二十二碳六烯酸(DHA)的脂肪酸乙酯与甘油进行酯交换反应合成甘油酯。在17种常用树脂中,环氧树脂(ECR8285)在缓冲液p H 5、缓冲液离子浓度1 mol/L、酶加量120mg/g(以载体质量计)时,通过共价结合固定化的LipozymeCALB L酶活力最高,可以达到22 967U/g,蛋白质吸附率为91.88%。以ECR8285固定化的LipozymeCALB L催化富含EPA和DHA的脂肪酸乙酯与甘油反应合成甘油酯,在叔丁醇体系中催化PUFA乙酯与甘油反应8 h后,PUFA乙酯的转化率可以达到51.1%;将叔丁醇回收后,在真空状态下继续反应24 h,PUFA乙酯的转化率可以达到91.7%,产物中甘油三酯的含量可以达到70%以上。Abstract:Lipozyme^(R) CALB L was immobilized by resin and was employed to synthesize triglycerides by transesterification of glycerol and EPA/DHA - enriched ethyl esters. Among commonly used seventeen kinds of resins, epoxy resin ECR8285 immobilized Lipozyme^(R) CALB L through covalent combination had the highest enzyme activity of 22 967 U/g, and its adsorption rate of protein was 91.88% under the condi- tions of buffer solution pH 5.0, ionic concentration of buffer solution 1 moL/L and dosage of enzyme 120 mg/g(based on the mass of carrier). Then ECR8285 immobilized Lipozyme^(R) CALB L was applied to syn- thesize triglycerides by transesterification of glycerol and EPA/DHA - enriched ethyl esters, and the con- version rate of PUFA ethyl esters reached 51.1% after transesterification of PUFA ethyl esters and glycerol for 8 h in tert - butanol system. And the conversion rate of PUFA ethyl esters and the content of triglycer- ides in product reached 91.7% and above 70% respectively when the transesterification continued for 24 h under vacuum after tert -butanol being recovered.
