摘　　要：目的检测急性髓系白血病(AML)中Galectin-1基因的甲基化水平,探讨Galectin-1基因表达水平与启动子甲基化的关系。方法选取2014年1月—2015年12月在该院门诊部治疗前AML骨髓54例、对照骨髓35例和治疗后完全缓解AML骨髓8例,采用采用甲基化特异性PCR(MS-PCR)方法检测Galectin-1基因甲基化状态。结果 Galectin-1基因在AML骨髓中的平均表达水平明显低于对照骨髓(0.35±0.09)VS(0.87±0.22),在完全缓解AML骨髓中明显高于治疗前骨髓(0.79±0.21)VS(0.33±0.11);Galectin-1基因在AML骨髓的甲基化阳性率明显高于对照骨髓(79.6%VS 17.1%),Galectin-1基因的甲基化阳性率在完全缓解AML骨髓中明显低于治疗前骨髓(12.5%VS 75.0%);Galectin-1基因表达水平与其甲基化呈负相关。结论Galectin-1基因在AML中出现表达下调或缺失,与其启动子甲基化有关。Objective To detect acute myeloid leukemia（AML） methylation status of Galectin-1 gene, to investigate the expression and promoter methylation. Methods Our out-patient department, 54 patients during January 2014 to December 2015 with AML before the treatment of bone marrow, bone marrow of 35 cases of the control and 8 cases of complete remission after treatment of AML bone mar-row samples by methylation specific PCR（MS-PCR） to detect the methylation status of Galectin-1 gene. Results Galectin-1 gene ex-pression levels in bone marrow in average AML was significantly lower than the control of bone marrow（0.35 ±0.09） VS（0.87±0.22） in complete remission, average expression level of AML in bone marrow was significantly higher than that before treatment of bone mar-row（0.79±0.21） VS（0.33±0.11）; Galectin-1 gene in bone marrow of methyl AML positive rate was significantly higher in the bone marrow（79.6% VS 17.1%）, in complete remission in the bone marrow of methyl AML positive rate was significantly lower than before treatment（12.5% VS 75%）; the expression level of Galectin-1 gene and its methylation negative correlation. Conclusion Galectin-1gene in AML table Up to down or missing, and its promoter methylation related.
