摘  要:目的:研究香菇多糖体外逆转A2780卵巢癌细胞顺铂耐药及可能机制。方法:根据药物处理方法将A2780卵巢癌细胞分为CON组(未行香菇多糖或顺铂处理)、DDP组(仅用顺铂处理)、LEN组(仅用香菇多糖处理)及D+L组(香菇多糖联合顺铂处理),比较四组细胞增殖、细胞周期及凋亡率和耐药基因mRNA表达的差异。结果:D+L组细胞第1-7天的吸光度、S期、G2/M期和PI均显著低于CON组、DDP组和LEN组细胞(均P<0.05),G0/G1期和细胞凋亡率均显著高于CON组、DDP组和LEN组细胞(均P<0.05);LEN组和D+L组细胞MDR1和MRP1基因mRNA表达无明显差异(均P>0.05),CON组和DDP组细胞MDR1和MRP1基因mRNA表达显著高于LEN组和D+L组细胞(均P<0.05)。结论:香菇多糖体外可逆转卵巢癌细胞对顺铂耐药性,可能与降低MDR1和MRP1表达有关。Objective:To study the reversing effect of lentinan on the resistance of A2780 ovarian cancer cellto Cisplatin in vitro and its mechanism.Methods:A2780ovarian cancer cells were divided into CON group(no Lentinan and Cisplatin treatment),DDP group(only Cisplatin treatment),LEN group(only lentinan treatment),and D+L group(both lentinan and Cisplatin treatment)according to their treatments.The differences of cell proliferation,cell cycle,apoptosis and expression of multi-drug resistance gene were compared.Results:The D1-D7 absorption,S stage,G2/M stage and PI in D+L group were significantly lower than those in CON group,DDP group and LEN group(P〈0.05),while G0/G1 stage and cell apoptosis rate were higher(P〈0.05).The differences of MDR1 and MRP1’s mRNA expression were not statistics significant between LEN group and D+L group(P〉0.05),while their expression were lower than those in CON group and DDP group(P〈0.05).Conclusion:Lentinan could reverse the resistance of A2780 ovarian cancer cell to Cisplatin in vitro via lowering down MDR1 and MRP1expression.
