胃癌组织Twist与MMP-2的表达及临床病理意义

摘　　要：目的:通过探讨Twist和基质金属蛋白酶2(MMP-2)在胃癌组织中的表达水平,以研究两者与胃癌临床病理因素的相互关系,从而为临床上胃癌的转移复发预警及预后评估提供更准确的研究靶点。方法:首先收集37例胃癌患者石蜡组织切片及其对应的临床病例资料,然后通过免疫组化的方法检测Twist和MMP-2在37例胃癌病理组织切片中的表达情况;通过统计分析进一步了解Twist和MMP-2与临床胃癌相关病理因素之间的关系,同时对两者之间表达水平的关联性进行分析。结果:Twist蛋白在肿瘤细胞的胞质中呈现高表达,同时也存在于细胞核中,其阳性表达率为56.8%;胃癌的淋巴结转移、浆膜浸润等主要与Twist基因的表达有关(P<0.05);MMP-2蛋白在肿瘤细胞的胞质中呈现高表达,同时也存在于细胞核中,其阳性表达率为54.1%;胃癌的复发及远处转移与MMP-2的表达有关(P<0.05);Twist和MMP-2在胃癌组织中的表达呈正相关(r=0.257,P<0.05)。结论:胃癌组织中Twist和MMP-2表达均存在异常,且两者的表达水平的变化与临床上胃癌的转移、复发等过程密切相关;同时在一定程度上胃癌组织中的Twist的表达水平可能会调控MMP-2的表达。Objective：To investigate the expression of Twist and matrix metalloproteinase-2（MMP-2）protein in gastric carcinoma and study their clinical pathological correlations in human gastric carcinoma as well as their relationships.Methods：A total of 37paraffin-embedded gastric carcinoma were studied by immunohistochemistry to evaluate the protein expression status of Twist and MMP-2protein.The relation was analyzed between the protein expression and clinical pathological conditions.Also the correlation of the two factors’ expression was examined.Results：Twist protein was mainly expressed in the cytoplasm of tumor cells,partly in the nuclei of tumor cells.The positive rate of Twist protein expression was 56.8%.The positive expression of Twist was positively correlated with serosal invasion and lymph node metastasis（P〈0.05respectively）.MMP-2protein was mainly expressed in the cytoplasm of tumor cells,and partly in the nuclei of tumor cells.The positive rate of MMP-2protein expression was 54.1%.And the positive expression of MMP-2was positively correlated with recurrence and distant metastasis（P〈0.05respectively）.The expression of MMP-2was positively correlated with Twist expression（r=0.257,P〈0.05）.Conclusion：The expression of Twist and MMP-2are positively correlated with the invasion and metastasis of gastric cancer;MMP-2expression is regulated by Twist at a certain level.
