摘 要:利用双水相萃取技术从牛乳乳清中提取乳过氧化物酶。以乳过氧化物酶酶活回收率和纯化倍数为指标,研究双水相体系的盐相、PEG分子质量、PEG浓度、硫酸镁含量及体系pH值对乳过氧化物酶分布在上相或下相的影响。研究结果表明,当双水相体系的pH值为8.5时,选择10%硫酸镁和16%PEG 6 000,可将乳过氧化物酶富集在盐相,乳过氧化物酶的酶活回收率和纯化倍数分别为153.59%和1.99倍。Lactoperoxidase was extracted and separated from bovine milk by coupling aqueous two-phase. Taking activity recovery of lactoperoxidase and purification factor as index, the effect of aqueous two-phase extraction system parameters such as type of phase system, polyethylene glycol(PEG) molecular mass, concentration of PEG, concentration of magnesium sulfate and system pH on lactoperoxidase in top and bottom phases was studied. The resuts showed that when there was pH 8.5, 10% magnesium sulfate and 16% PEG 6000 in aqueous two-phase extraction system, lactoperoxidase gathered in the phase of salt. And its lactoperoxidase activity recovery and purification factor were 153.59% and1.99 times, respectively.
