摘 要:目的:研究AG1478对宫颈癌HeLa细胞凋亡的影响,为宫颈癌治疗提供初步理论基础。方法:通过CCK8、Western blot、RT-qPCR、免疫荧光及TUNEL染色,检测AG1478对HeLa细胞增殖的影响;凋亡相关基因及蛋白表达的改变;磷酸化ERK亚细胞定位的分布及早期凋亡现象。结果:AG1478对HeLa细胞的增殖有抑制作用;对EGFR、ERK及AKT的磷酸化活性有抑制作用同时激活凋亡信号通路;磷酸化ERK细胞核转位被阻断同时出现早期凋亡现象。结论:AG1478可以通过抑制EGFR及下游PI3K/AKT及Ras/MAPK信号通路抑制HeLa细胞增殖并引起凋亡。Objective To investigate the biological features of epidermal growth factor receptor inhibitorAG1478 on cervical carcinoma cell. Methods The proliferation of He La cells under AG1478 stimulation wasdetermined by CCK8 assay. The expression of EGFR downstream signaling protein and apoptotic relative proteinwere examined by Western blot and transcription of apotosis-related genes were measured by RT-q PCR in AG1478 treated He La cells. Nuclear transport of phosphorylated ERK were measured through ICC assay. TUNEL assay wasused to determine early stage of apoptosis. Results CCK8 assay showed that AG1478 inhibit the proliferation ofHe La cells and also block phosphorylated level of EGFR,ERK and AKT. Furthermore,nuclear transport ofphosphorylated ERK upon EGF stimulation were blocked and pro-apoptotic proteins were up-regluated with activatedcleaved Caspases. Conclusion AG1478 inhibited the proliferation and induced apoptosis in He La cells and itcould be a potential therapeutic drug for cervical carcinoma.
