摘 要:在低能N+注入沙漠寡营养细菌DOB150菌株获得重组突变菌株DOB981的基础上,本研究应用生物信息学方法从DOB全基因组De novo数据中获得了nif H基因信息,分析结果表明重组突变菌株DOB981的2个拷贝nif H基因中分别有39个和5个碱基位点发生了突变。通过分析nifH基因的蛋白质二级结构,获得了蛋白质的结构域、功能位点、跨膜区域和卷曲螺旋等基本信息,为进一步理解低能离子注入介导的功能基因突变提供了分子证据。The original desert oligotrophic bacteria strain DOB150 was implanted by low-energy nitrogen ion (N^+) to obtain the recombinant mutant strain DOB981 in the preliminary study, and n/fH genetic information were obtained from DOB genome De novo data by bioinformatics methods in this study. The results of analysis showed that there were 39 and 5 base sites mutated in two copies nifH genes of the recombinant mutant strain DOB981. The structure of the protein domain, functional site, the trans-membrane region, the coiled-coil and other basic information were obtained by analyzing the protein secondary structure of nifH gene, which provided the molecular evidence for further understanding of functional gene mutation mediated by low energy ion implantation.
