摘 要:研究蓝莓花色苷对东莨菪碱诱导小鼠学习记忆障碍的影响,并初步探讨其作用机制。雄性KM种小鼠60只,随机分为6组,分别为空白对照组、模型对照组、阳性对照组(吡拉西坦片200 mg/kg·d)、蓝莓花色苷高剂量组(400 mg/kg·d)、蓝莓花色苷中剂量组(200 mg/kg·d)、蓝莓花色苷低剂量组(100 mg/kg·d),各组连续给药14 d,从实验第15天开始,除空白对照组外,其余各试验组均注射东莨菪碱2 mg/kg,建立小鼠记忆障碍模型,末次给药2 h后,进行跳台试验训练。测试完毕,断头处死小鼠,检测蓝莓花色苷对脑组织乙酰胆碱酯酶(Ach E)活性的影响,免疫印迹法检测海马组织NF-κB p65和Cleaved caspase-3的影响。结果表明:蓝莓花色苷各剂量组能够明显延长小鼠跳台潜伏期,减少5 min内出现的错误次数(P<0.05),且可以降低小鼠脑组织海马及皮层ACh E活性(P<0.05)。免疫印迹结果表明,蓝莓花色苷能够降低东莨菪碱诱导小鼠海马组织NF-κB p65和Cleaved caspase-3蛋白的表达。蓝莓花色苷对东莨菪碱模型小鼠的学习记忆能力具有一定的改善作用,其机制可能通过影响NF-k B信号转导通路而抑制东莨菪碱诱导的氧化应激和炎性反应。To observe the effect of blueberry anthocyanins on scopolamine-induced learning and memory impairment in mice. Sixty male KM mice were randomly divided into 6 groups:a blank control group,a model control group,a positive control group(laci staw 200 mg/kg·d),a high-dose blueberry anthocyanins group(400 mg/kg·d),a middle-dose blueberry anthocyanins group(200 mg/kg·d)and a low-dose blueberry anthocyanins group(100 mg/kg·d). Each group was continuously administered 14 days. On the 15 th day,except for the mice in blank group,other mice were injected scopolamine 2 mg/kg to establish memory impairment model.The step-down passive avoidance test was performed after the last administration of 2 hours. After testing,all mice were beheaded to detect the effect of Ach E. The expression of NF-κB p65 and Cleaved caspase-3 in the hippocampus of mice was detected by Western blotting. The results showed that:the blueberry anthocyanins in each dose group could significantly increase the incubation period platform in mice,decreased the mistaken reaction times in 5 min(P〈0.05),and could reduce ACh E activity in the cerebral cortex and hippocampus(P〈0.05). The result of Western blotting showed that blueberry anthocyanins could significantly reduce the expression of NF-κB p65 and Cleaved caspase-3 protein in the hippocampus of scopolamine-induced mice. Blueberry anthocyanins have certain improving effects on memory impairment induce by scopolamine in mice model of learning and memory disorder.
