摘 要:选取银杏叶片为试验材料,研究纤维素酶Umcel9y-1在银杏叶片总黄酮提取中的应用价值。试验结果表明,0.08 mg·mL^(-1)纤维素酶Umcel9y-1,在其最适反应条件下(pH7.0,37oC),处理银杏叶片1 h、2 h、4 h和过夜(12 h),总黄酮的提取量较对照组分别提高了20.3%、20.7%、16.8%和15.0%。纤维素酶试验组处理2 h,总黄酮的提取量可以达到对照组12 h提取所得总黄酮量的97.0%,纤维素酶Umcel9y-1显著缩短了银杏叶总黄酮提取时间。采用超高效液相色谱对2种提取方法获得的提取液进行药用成分检测,发现添加纤维素酶Umcel9y-1对中药提取液活性成分没有显著的影响。Ginkgo leaves were selected as experimental materials to investigate the cellulase Umcel9y-1 syner- gized action on total flavonoids extraction. Under the optimum reaction conditions (pH 7.0, 37℃), the extraction yields of total flavonoids increased by 20.3%, 20.7 %, 16.8% and 15% of the corresponding control after the leaves were treated with 0.08 mg.mL-1 Umcel9y-1 for 1 h, 2 h, 4 h and overnight (12 h). The extraction yield oftoal flavon- oids in the 2-hour treatment group reached 97.01% of the overnight-treatment of the control. The results showed the extract efficiency was greatly improved by the addition of cellulase Umcel9y-1. Moreover, the medical ingredients of the extracts were evaluated by Ultra performance liquid chromatography (UPLC), indicating that Umcel9y-1 had no obvious influence on the medical ingredients of the extract compared to the corresponding control.
