摘  要:叶绿素a是反映水体中浮游植物生物量的重要指标,其含量的准确测定具有十分重要的意义.以实验室培养的铜绿微囊藻为例,比较了常用的四种提取方法和三种测量方法对叶绿素a浓度测量结果的影响.实验结果表明:使用热丙酮法提取叶绿素a浓度效率最高;不同测量方法得到的叶绿素a浓度结果,分光光度计三色法>分光光度计单色法>荧光法.测量方式不同对叶绿素a浓度结果的影响大于提取方式的不同对浓度结果的影响,并且不同提取或者测量方法得到的结果之间具有极好的线性关系.Chlorophyll a is an important indicator of phytoplankton biomass in water, and accurate determination of its con-tent is of great significance. Taking Microcystis aeruginosa cultivated in the laboratory as the research object, this paper compares four different extraction methods and three different measurements used currently. Experimental results show that chlorophyll a concentration extracted by hot ethanol method is most effective, and chlorophyll a concentration measured by different methods, three-color spectrophotometry 〉 single-color spectrophotometry 〉 fluorescence method. Measurement difference has more effects on the chlorophyll a concentration result than extraction difference, and chlorophyll a concentration extracted or measured by different methods has high linear relationship.
